#133 Immunological and pathophysiological characteristics <str<strong>on</strong>g>of</str<strong>on</strong>g> Mycobacterium avium<br />

subspecies paratuberculosis infecti<strong>on</strong> in guinea pigs<br />

In order to elucidate <str<strong>on</strong>g>the</str<strong>on</strong>g> subject why pr<str<strong>on</strong>g>of</str<strong>on</strong>g>ound chr<strong>on</strong>ic enteritis is caused by Mycobacterium avium subspecies<br />

paratuberculosis (Map) in ruminants but not by o<str<strong>on</strong>g>the</str<strong>on</strong>g>r M. avium subspecies which have great similarity in<br />

genetical and antigenical characteristics, we have tried to find differences that may characterize Map infecti<strong>on</strong><br />

from those <str<strong>on</strong>g>of</str<strong>on</strong>g> o<str<strong>on</strong>g>the</str<strong>on</strong>g>r mycobcaterial infecti<strong>on</strong>s.<br />

In preliminary experiments using guinea pigs, shedding <str<strong>on</strong>g>of</str<strong>on</strong>g> intraperit<strong>on</strong>eally inoculated Mycobacterium<br />

organisms into feces was detected with surprising speed <str<strong>on</strong>g>of</str<strong>on</strong>g> within 24 hours after inoculati<strong>on</strong>, <str<strong>on</strong>g>the</str<strong>on</strong>g>refore <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

experiments were divided into two different observati<strong>on</strong> periods <str<strong>on</strong>g>of</str<strong>on</strong>g> 24 hours and two m<strong>on</strong>ths after inoculati<strong>on</strong>.<br />

Four-week-old female Hartley guinea pigs were intraperit<strong>on</strong>eally inoculated with 5 different species or<br />

subspecies <str<strong>on</strong>g>of</str<strong>on</strong>g> live mycobacterium (Map, M. avium subspecies avium, M. intracellulare M. scr<str<strong>on</strong>g>of</str<strong>on</strong>g>ulaceum, and<br />

M. bovis). After 24 hours or two m<strong>on</strong>ths, whole intestine and principal organs were collected and performed<br />

bacteriological and histopathological examinati<strong>on</strong>s with bacterial culture, quantificati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> bacteria by realtime<br />

PCR and immuno-pathological staining using anti CD68 m<strong>on</strong>ocl<strong>on</strong>al antibody.<br />

Results obtained from our experiments are as follows: 1) Comm<strong>on</strong> findings am<strong>on</strong>g all Mycobacterial<br />

infecti<strong>on</strong>s: Granuloma formati<strong>on</strong> in liver and spleen; Thickening <str<strong>on</strong>g>of</str<strong>on</strong>g> duodenum because <str<strong>on</strong>g>of</str<strong>on</strong>g> infiltrati<strong>on</strong> with inflammatory<br />

cells; Bacterial shedding within 24 hours after intraperit<strong>on</strong>eal inoculati<strong>on</strong>. 2) Characteristics in Map and<br />

o<str<strong>on</strong>g>the</str<strong>on</strong>g>r M. avium subspecies infecti<strong>on</strong>s: Jejunoileal inflammati<strong>on</strong> c<strong>on</strong>sisted <str<strong>on</strong>g>of</str<strong>on</strong>g> macrophages, plasma cells, and<br />

eosinophils at 2 m<strong>on</strong>ths postinoculati<strong>on</strong>. 3) Characteristics exclusively observed in Map infecti<strong>on</strong>: Significantly<br />

lower number <str<strong>on</strong>g>of</str<strong>on</strong>g> organisms in <str<strong>on</strong>g>the</str<strong>on</strong>g> duodenum at 24 hours after inoculati<strong>on</strong> than those <str<strong>on</strong>g>of</str<strong>on</strong>g> o<str<strong>on</strong>g>the</str<strong>on</strong>g>rs; - Scattered distributi<strong>on</strong><br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> CD68 + macrophages in <str<strong>on</strong>g>the</str<strong>on</strong>g> lamina propria mucosae <str<strong>on</strong>g>of</str<strong>on</strong>g> duodenal villi c<strong>on</strong>trary to focused distributi<strong>on</strong><br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> those cells at <str<strong>on</strong>g>the</str<strong>on</strong>g> tip <str<strong>on</strong>g>of</str<strong>on</strong>g> villi in o<str<strong>on</strong>g>the</str<strong>on</strong>g>r Mycobacterial infecti<strong>on</strong>s.<br />

These observati<strong>on</strong>al data generated from guinea pig infecti<strong>on</strong>s may help us to understand <str<strong>on</strong>g>the</str<strong>on</strong>g> pathogenesis<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> Map infecti<strong>on</strong>.<br />
