The way we approach <str<strong>on</strong>g>the</str<strong>on</strong>g> study <str<strong>on</strong>g>of</str<strong>on</strong>g> molecular pathogenesis <str<strong>on</strong>g>of</str<strong>on</strong>g> M. paratuberculosis, <str<strong>on</strong>g>the</str<strong>on</strong>g> causative agent <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

Johne’s disease in cattle, has changed dramatically with <str<strong>on</strong>g>the</str<strong>on</strong>g> arrival <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> genomics era by sequencing <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

genome <str<strong>on</strong>g>of</str<strong>on</strong>g> M. paratuberculosis. My group am<strong>on</strong>g o<str<strong>on</strong>g>the</str<strong>on</strong>g>r groups led <str<strong>on</strong>g>the</str<strong>on</strong>g> way in developing novel tools to<br />

dissect <str<strong>on</strong>g>the</str<strong>on</strong>g> genetic basis <str<strong>on</strong>g>of</str<strong>on</strong>g> M. ap infecti<strong>on</strong>s <strong>on</strong> a genome-wide scale. One <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> employed approaches was<br />

<str<strong>on</strong>g>the</str<strong>on</strong>g> c<strong>on</strong>structi<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> mutant library <str<strong>on</strong>g>of</str<strong>on</strong>g> M. ap that was screened to identify genes important for virulence as well<br />

as for lipid biosyn<str<strong>on</strong>g>the</str<strong>on</strong>g>sis. Ano<str<strong>on</strong>g>the</str<strong>on</strong>g>r approach involved <str<strong>on</strong>g>the</str<strong>on</strong>g> development <str<strong>on</strong>g>of</str<strong>on</strong>g> DNA microarrays to analyze <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

transcripti<strong>on</strong>al pr<str<strong>on</strong>g>of</str<strong>on</strong>g>ile <str<strong>on</strong>g>of</str<strong>on</strong>g> both <str<strong>on</strong>g>the</str<strong>on</strong>g> bovine host and <str<strong>on</strong>g>the</str<strong>on</strong>g> infecting M. ap growing under variable c<strong>on</strong>diti<strong>on</strong>s. Finally,<br />

sequence-based approaches were introduced to investigate genomic compositi<strong>on</strong> and genomic topology <str<strong>on</strong>g>of</str<strong>on</strong>g> M.<br />

ap. In this presentati<strong>on</strong>, I will discuss <str<strong>on</strong>g>the</str<strong>on</strong>g> latest <strong>on</strong> <str<strong>on</strong>g>the</str<strong>on</strong>g> pathogenesis <str<strong>on</strong>g>of</str<strong>on</strong>g> M. ap infecti<strong>on</strong> following <str<strong>on</strong>g>the</str<strong>on</strong>g> applicati<strong>on</strong><br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> key genomic approaches. Our goal here is to shed lights <strong>on</strong> <str<strong>on</strong>g>the</str<strong>on</strong>g> current and future efforts that will shape <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

field <str<strong>on</strong>g>of</str<strong>on</strong>g> Johne’s disease to better understand <str<strong>on</strong>g>the</str<strong>on</strong>g> pathogenesis <str<strong>on</strong>g>of</str<strong>on</strong>g> this disease and to develop novel interventi<strong>on</strong><br />

strategies against Johne’s disease.<br />
