#16 Histopathological and immunohistochemical studies <strong>on</strong> naturally occurring<br />

paratuberculosis in goats<br />

Immunohistochemical and histopathological studies were carried out <strong>on</strong> sp<strong>on</strong>taneously occurring paratuberculosis<br />

(Johne’s disease) in 38 goats. Am<strong>on</strong>g <str<strong>on</strong>g>the</str<strong>on</strong>g>se goats, focal (grade 1), diffuse lymphocytic (grade 2), diffuse<br />

mixed (grade 3) and diffuse multibacillary (grade 4) lesi<strong>on</strong>s were observed in 3(7.8%), 5(13.1%), 15(39.4%),<br />

15(39.4%) goats, respectively. The focal lesi<strong>on</strong>s, tiny granulomas were observed in <str<strong>on</strong>g>the</str<strong>on</strong>g> interfollicular areas <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

Peyer’s patches (PP), and <str<strong>on</strong>g>the</str<strong>on</strong>g> cortex <str<strong>on</strong>g>of</str<strong>on</strong>g> mesenteric lymph node (MLN). In diffuse lymphocytic lesi<strong>on</strong>s, besides<br />

<str<strong>on</strong>g>the</str<strong>on</strong>g> presence <str<strong>on</strong>g>of</str<strong>on</strong>g> granulomas in <str<strong>on</strong>g>the</str<strong>on</strong>g> Peyer’s patches, jejunal and ileal mucosae were diffusely infiltrated with<br />

lymphoid cells and small number <str<strong>on</strong>g>of</str<strong>on</strong>g> macrophages. Small multifocal granulomas were observed in <str<strong>on</strong>g>the</str<strong>on</strong>g> cortex<br />

and paracortex <str<strong>on</strong>g>of</str<strong>on</strong>g> MLN. Goats with diffuse mixed and diffuse multibacillary lesi<strong>on</strong>s had shown poor body c<strong>on</strong>diti<strong>on</strong>s<br />

and thickened and corrugated intestinal mucosae <strong>on</strong> necropsy. Histologically, in diffuse mixed lesi<strong>on</strong>s,<br />

mucosa showed infiltrati<strong>on</strong> with epi<str<strong>on</strong>g>the</str<strong>on</strong>g>lioid cells mixed with lymphocytes. Small to large multifocal granulomas<br />

c<strong>on</strong>taining clusters <str<strong>on</strong>g>of</str<strong>on</strong>g> acid-fast bacilli (AFB) were observed in <str<strong>on</strong>g>the</str<strong>on</strong>g> MLN. In diffuse multibacillary lesi<strong>on</strong>s, intestinal<br />

jejunal and ileal villi were club shaped, at times flat and fused, and diffusely infiltrated with sheets <str<strong>on</strong>g>of</str<strong>on</strong>g> epi<str<strong>on</strong>g>the</str<strong>on</strong>g>lioid<br />

macrophages loaded with clusters <str<strong>on</strong>g>of</str<strong>on</strong>g> AFB. In indirect immunoperoxidase test (IPT), 36 (94.7%) out <str<strong>on</strong>g>of</str<strong>on</strong>g> 38<br />

goats showed positive immunoreacti<strong>on</strong>, <str<strong>on</strong>g>the</str<strong>on</strong>g> intensity <str<strong>on</strong>g>of</str<strong>on</strong>g> which varied with <str<strong>on</strong>g>the</str<strong>on</strong>g> types <str<strong>on</strong>g>of</str<strong>on</strong>g> lesi<strong>on</strong>s. On comparis<strong>on</strong>,<br />

<str<strong>on</strong>g>the</str<strong>on</strong>g> IPT was found to have better sensitivity than Ziehl-Neelsen (ZN) staining (84.2%). One <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> most important<br />

advantages <str<strong>on</strong>g>of</str<strong>on</strong>g> IPT was that <str<strong>on</strong>g>the</str<strong>on</strong>g> positive immunoreacti<strong>on</strong> could be visualized even at low magnificati<strong>on</strong> in<br />

all grades <str<strong>on</strong>g>of</str<strong>on</strong>g> lesi<strong>on</strong>s including focal and diffuse lymphocytic type, in which AFB were scarcely detected with<br />

oil immersi<strong>on</strong> objective. Based <strong>on</strong> this study, it was c<strong>on</strong>cluded that a spectrum <str<strong>on</strong>g>of</str<strong>on</strong>g> pathology existed in caprine<br />

paratuberculosis. IPT was more sensitive than ZN for dem<strong>on</strong>strati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> MAP bacilli/antigens.<br />
