Map-exposed heifer calves: a field study<br />

Johne’s disease is present in a high percentage <str<strong>on</strong>g>of</str<strong>on</strong>g> dairy herds and causes major ec<strong>on</strong>omic losses to <str<strong>on</strong>g>the</str<strong>on</strong>g> dairy<br />

industry. Diagnostic tests for detecting serum antibody are not <str<strong>on</strong>g>of</str<strong>on</strong>g> suitable sensitivity for m<strong>on</strong>itoring M. avium ss<br />

paratuberculosis (Map) infecti<strong>on</strong> in young replacement cattle or for detecting subclinical infecti<strong>on</strong> in cattle. The<br />

specific objectives were to: 1. Obtain informati<strong>on</strong> <strong>on</strong> skin test resp<strong>on</strong>ses in heifers from Map positive herds,<br />

and 2. Determine <str<strong>on</strong>g>the</str<strong>on</strong>g> specificity <str<strong>on</strong>g>of</str<strong>on</strong>g> skin tests using Map PPD in heifers 10 to 26 m<strong>on</strong>ths <str<strong>on</strong>g>of</str<strong>on</strong>g> age.<br />

The MAP-PPD used in this investigati<strong>on</strong> was prepared by precipitati<strong>on</strong> with trichloroacetic acid (TCA)<br />

from <str<strong>on</strong>g>the</str<strong>on</strong>g> culture filtrate <str<strong>on</strong>g>of</str<strong>on</strong>g> Mycobacterium avium ss paratuberculosis ATCC Neotype Strain 19698. The skin test<br />

was c<strong>on</strong>ducted in <str<strong>on</strong>g>the</str<strong>on</strong>g> mid-cervical regi<strong>on</strong>; skin thickness measured (mm) before injecti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> PPD and 72 hours<br />

following injecti<strong>on</strong>. Skin tests were c<strong>on</strong>ducted in 842 heifers were from 10 herds in which Johne’s disease had<br />

been diagnosed by mycobacteriologic examinati<strong>on</strong> and in 468 heifers <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> same age which were from 16<br />

herds in which Johne’s disease had not been diagnosed.<br />

The results revealed that positive skin test resp<strong>on</strong>ses were detected in heifers 10 to 26 m<strong>on</strong>ths <str<strong>on</strong>g>of</str<strong>on</strong>g> age<br />

in each <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> 10 Johne’s positive herds; positive ELISA reacti<strong>on</strong>s were detected in heifers in <strong>on</strong>ly 3 <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> 10<br />

Johne’s positive herds. The specificity <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> skin test using Map was 98% in heifers 10 to 26 m<strong>on</strong>ths <str<strong>on</strong>g>of</str<strong>on</strong>g> age,<br />

95% in cows 26 to 38 m<strong>on</strong>ths <str<strong>on</strong>g>of</str<strong>on</strong>g> age and 97% in cows 38 to 50 m<strong>on</strong>ths <str<strong>on</strong>g>of</str<strong>on</strong>g> age.<br />

Beside <str<strong>on</strong>g>the</str<strong>on</strong>g> <strong>on</strong>going discussi<strong>on</strong>s about <str<strong>on</strong>g>the</str<strong>on</strong>g> improvement <str<strong>on</strong>g>of</str<strong>on</strong>g> laboratory procedures, <str<strong>on</strong>g>the</str<strong>on</strong>g> genome <str<strong>on</strong>g>of</str<strong>on</strong>g> MAP and<br />

o<str<strong>on</strong>g>the</str<strong>on</strong>g>r important scientific topics a supraregi<strong>on</strong>al discussi<strong>on</strong> about a basically <strong>on</strong> farm program against paratuberculosis<br />

should be started.<br />

Many different countries have established voluntary programs to c<strong>on</strong>trol paratuberculosis and prevent<br />

fur<str<strong>on</strong>g>the</str<strong>on</strong>g>r spreading <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> disease. In o<str<strong>on</strong>g>the</str<strong>on</strong>g>r countries compulsory registrati<strong>on</strong> for paratuberculosis is performed,<br />

or strict c<strong>on</strong>trol and stamping out programs are in acti<strong>on</strong>.<br />

To start <str<strong>on</strong>g>the</str<strong>on</strong>g> discussi<strong>on</strong> and try to find agreement <strong>on</strong> cost effective and easy to perform steps to fight paratuberculosis,<br />

we would like to perform an expert-survey during <str<strong>on</strong>g>the</str<strong>on</strong>g> <str<strong>on</strong>g>10th</str<strong>on</strong>g> ICP <strong>on</strong> basically management suggesti<strong>on</strong>s<br />

for MAP-positive farms. We suggest that <str<strong>on</strong>g>the</str<strong>on</strong>g> survey is introduced in a short presentati<strong>on</strong> at <str<strong>on</strong>g>the</str<strong>on</strong>g> beginning<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> c<strong>on</strong>gress and carried out during brakes and poster sessi<strong>on</strong>s.<br />

The aim <str<strong>on</strong>g>of</str<strong>on</strong>g> this survey is to generate effective and simple measurements to reduce <str<strong>on</strong>g>the</str<strong>on</strong>g> incidence <str<strong>on</strong>g>of</str<strong>on</strong>g> paratuberculosis<br />

in affected herds. This should be <str<strong>on</strong>g>the</str<strong>on</strong>g> possible start <str<strong>on</strong>g>of</str<strong>on</strong>g> a trans-nati<strong>on</strong>al basic program in <str<strong>on</strong>g>the</str<strong>on</strong>g> fight<br />

against paratuberculosis and <str<strong>on</strong>g>the</str<strong>on</strong>g> protecti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> free herds and areas which could be implemented immediately.<br />

Beside all differences in acti<strong>on</strong>s and opini<strong>on</strong>s this could be c<strong>on</strong>sidered as a “minimum level” which could<br />

be achieved in reas<strong>on</strong>able time and would be accepted by many countries.<br />

Results <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> survey could be presented in <str<strong>on</strong>g>the</str<strong>on</strong>g> IAP <strong>Paratuberculosis</strong> Newsletter.<br />
