stored at -20ºC until fur<str<strong>on</strong>g>the</str<strong>on</strong>g>r analysis. The antigen specific IFN-� producti<strong>on</strong> in supernatants<br />

were determined by an in-house ELISA as described elsewhere (Mikkelsen et al., 2009). The level<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> IFN-� (pg/ml) was calculated using linear regressi<strong>on</strong> <strong>on</strong> log-log transformed reading from <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

two-fold diluti<strong>on</strong> series <str<strong>on</strong>g>of</str<strong>on</strong>g> a reference standard with known IFN-� c<strong>on</strong>centrati<strong>on</strong>.<br />

Samples were excluded if <str<strong>on</strong>g>the</str<strong>on</strong>g> IFN-� value in <str<strong>on</strong>g>the</str<strong>on</strong>g> SEB-stimulated sample was below 1500 pg/ml or<br />

<str<strong>on</strong>g>the</str<strong>on</strong>g> PBS-stimulated value was higher than 250 pg/ml. The correlati<strong>on</strong>s between IFN-� resp<strong>on</strong>ses<br />

in <str<strong>on</strong>g>the</str<strong>on</strong>g> three types <str<strong>on</strong>g>of</str<strong>on</strong>g> samples and <strong>on</strong> different sampling dates were <str<strong>on</strong>g>the</str<strong>on</strong>g>n investigated.<br />

RESULTS<br />

In Figure 1 <str<strong>on</strong>g>the</str<strong>on</strong>g> IFN-� (pg/ml) resp<strong>on</strong>ses to PPDj-stimulati<strong>on</strong> in fresh samples and day-old samples<br />

+ IL-12 from all 3 sampling dates are compared. The greatest correlati<strong>on</strong> was observed at <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

first sample date (R 2 = 0.72) whereas lower correlati<strong>on</strong>s were observed at <str<strong>on</strong>g>the</str<strong>on</strong>g> sec<strong>on</strong>d (R 2 =<br />

0.16) and third (R 2 = 0.21) sample date.<br />

Again, <str<strong>on</strong>g>the</str<strong>on</strong>g> greatest correlati<strong>on</strong> was observed at <str<strong>on</strong>g>the</str<strong>on</strong>g> first sample date (R 2 = 0.67), and lower<br />

correlati<strong>on</strong>s at <str<strong>on</strong>g>the</str<strong>on</strong>g> sec<strong>on</strong>d (R 2 = 0.09) and third (R 2 = 0.35) sample date.<br />

The highest correlati<strong>on</strong>s were observed between fresh samples and day-old samples<br />

potentiated with IL-12, but <str<strong>on</strong>g>the</str<strong>on</strong>g>re was little difference between additi<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> IL-12 and anti-IL-10<br />

antibodies. The between-sample day variati<strong>on</strong> was high for both types <str<strong>on</strong>g>of</str<strong>on</strong>g> day-old samples<br />

irrespective <str<strong>on</strong>g>of</str<strong>on</strong>g> treatment type. We have previously observed fluctuating IFN-� resp<strong>on</strong>ses in<br />
