Missouri). Serum from a cow with documented Johne’s disease c<strong>on</strong>stituted <str<strong>on</strong>g>the</str<strong>on</strong>g> positive<br />

c<strong>on</strong>trol. Final analytical readings were d<strong>on</strong>e at 24 and 48 hours. The appearance <str<strong>on</strong>g>of</str<strong>on</strong>g> <strong>on</strong>e or<br />

more clearly definable precipitati<strong>on</strong> lines before or at 48 hours c<strong>on</strong>stituted a positive result.<br />

The AGID tests were d<strong>on</strong>e with both n<strong>on</strong>absorded and Mycobacterium phlei absorbed sera.<br />

Pre-absorbed ELISA test: Test sera were pre-absorbed with M. phlei. The ELISA results<br />

were calculated from wavelength readings at optical density (OD) 405 nm. All readings less<br />

than 1.6 OD were c<strong>on</strong>sidered negative; readings between 1.5 and 1.9 OD were deemed<br />

suspicious/inc<strong>on</strong>clusive; and readings <str<strong>on</strong>g>of</str<strong>on</strong>g> 2.0 to 2.5 OD were called low positive. A high<br />

positive was any reading 2.6 OD or above.<br />

RESULTS<br />

Frequency <str<strong>on</strong>g>of</str<strong>on</strong>g> double precipitati<strong>on</strong> bands within <str<strong>on</strong>g>the</str<strong>on</strong>g> AGID test: Of <str<strong>on</strong>g>the</str<strong>on</strong>g> 71 individual animals<br />

identified having a positive AGID test, 13 (18%) exhibited, at <strong>on</strong>e point or ano<str<strong>on</strong>g>the</str<strong>on</strong>g>r, a double<br />

immunoprecipitati<strong>on</strong> band.<br />

Relati<strong>on</strong>ship <str<strong>on</strong>g>of</str<strong>on</strong>g> double precipitati<strong>on</strong> bands to corresp<strong>on</strong>ding ELISA optical density readings:<br />

The mean ELISA optical density reading (OD) for <str<strong>on</strong>g>the</str<strong>on</strong>g> 13 cows whose AGID test<br />

dem<strong>on</strong>strated a double precipitati<strong>on</strong> band was 3.88 with a range from 2.3 to 5.8. Only <strong>on</strong>e<br />

serum had an OD reading <str<strong>on</strong>g>of</str<strong>on</strong>g> less than 2.6.<br />

Single, primary precipitati<strong>on</strong> bands were not tightly correlated with <str<strong>on</strong>g>the</str<strong>on</strong>g> presence <str<strong>on</strong>g>of</str<strong>on</strong>g> a<br />

diagnostic Map ELISA test. Nine <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> 71 (12.7%) cases in which a double band occurred<br />

had a negative UF ELISA test. Ano<str<strong>on</strong>g>the</str<strong>on</strong>g>r 10 (14%) sera were deemed to be, at best,<br />

suspicious (Table 1). The greatest number <str<strong>on</strong>g>of</str<strong>on</strong>g> cases correlated with str<strong>on</strong>gly positive Map<br />

ELISA tests 39/71 (54.9%). Serial observati<strong>on</strong> in three cows dem<strong>on</strong>strated <str<strong>on</strong>g>the</str<strong>on</strong>g> unmasking <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

a sec<strong>on</strong>d immunoprecipitati<strong>on</strong> band when <str<strong>on</strong>g>the</str<strong>on</strong>g> corresp<strong>on</strong>ding ELISA titer became markedly<br />

elevated above that value at which <str<strong>on</strong>g>the</str<strong>on</strong>g> primary immunoprecipitati<strong>on</strong> band occurred (Table 2)<br />

When AGID positive sera were absorbed against varying c<strong>on</strong>centrati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> M. plea, <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

results for <str<strong>on</strong>g>the</str<strong>on</strong>g> immunoprecipitati<strong>on</strong> bands remained unchanged.<br />

DISCUSSION<br />

The comm<strong>on</strong> absence <str<strong>on</strong>g>of</str<strong>on</strong>g> an immunoprecipitati<strong>on</strong> band in <str<strong>on</strong>g>the</str<strong>on</strong>g> face <str<strong>on</strong>g>of</str<strong>on</strong>g> a high Map ELISA test<br />

result has been largely ascribed to <str<strong>on</strong>g>the</str<strong>on</strong>g> amount <str<strong>on</strong>g>of</str<strong>on</strong>g> specific antibody required for an<br />

immunoprecipitati<strong>on</strong> band in c<strong>on</strong>trast to <str<strong>on</strong>g>the</str<strong>on</strong>g> significantly smaller amount <str<strong>on</strong>g>of</str<strong>on</strong>g> specific antibody<br />

that is required for a diagnostic Map ELISA reading.<br />

Unlike <str<strong>on</strong>g>the</str<strong>on</strong>g> primary immunoprecipitati<strong>on</strong> band, <str<strong>on</strong>g>the</str<strong>on</strong>g> sec<strong>on</strong>d immunoprecipitati<strong>on</strong> band appears<br />

to have a positive correlati<strong>on</strong> with markedly elevated Map ELISA test readings. This<br />

observati<strong>on</strong> implies that <str<strong>on</strong>g>the</str<strong>on</strong>g> sec<strong>on</strong>d immunoprecipitati<strong>on</strong> band identifies predominantly <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

same Map antigen complexes that are used in <str<strong>on</strong>g>the</str<strong>on</strong>g> Map ELISA test.<br />

The resultant interpretati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> this data was that a better Map ELISA test could be developed<br />

using selected antigenic subsets embedded in <str<strong>on</strong>g>the</str<strong>on</strong>g> primary immunoprecipitati<strong>on</strong> band.<br />
