ABSTRACT<br />

The aim <str<strong>on</strong>g>of</str<strong>on</strong>g> this study was to determine <str<strong>on</strong>g>the</str<strong>on</strong>g> age structure <str<strong>on</strong>g>of</str<strong>on</strong>g> paratuberculosis infecti<strong>on</strong>s in<br />

culled dairy cattle as a mean to improve <str<strong>on</strong>g>the</str<strong>on</strong>g> design and use <str<strong>on</strong>g>of</str<strong>on</strong>g> diagnostic tests and c<strong>on</strong>trol<br />

strategies.<br />

581 adult Friesian cattle slaughtered in <str<strong>on</strong>g>the</str<strong>on</strong>g> Basque Country from March 2007 to<br />

December 2008 were included in <str<strong>on</strong>g>the</str<strong>on</strong>g> study. Samples <str<strong>on</strong>g>of</str<strong>on</strong>g> blood, intestinal tissue and<br />

mesenteric lymph nodes were taken from each animal. Blood was used in an ELISA test<br />

whereas intestinal tissue was used for bacterial isolati<strong>on</strong>, DNA detecti<strong>on</strong> by a commercial<br />

real time PCR (RTi-PCR) and histopathological examinati<strong>on</strong>. All methods showed a peak at<br />

3-4 years <str<strong>on</strong>g>of</str<strong>on</strong>g> age that ranged from 42.3% (histopathology) to 16.5% (ELISA). Minimum<br />

prevalence was found at 7-8 years. Overall prevalence ranged from 8.3% (ELISA) to 20.0%<br />

(RTi-PCR). Sampling within <str<strong>on</strong>g>the</str<strong>on</strong>g> 3-5 years <str<strong>on</strong>g>of</str<strong>on</strong>g> age group could improve <str<strong>on</strong>g>the</str<strong>on</strong>g> chances <str<strong>on</strong>g>of</str<strong>on</strong>g> herd<br />

infecti<strong>on</strong> detecti<strong>on</strong> with a minimum number <str<strong>on</strong>g>of</str<strong>on</strong>g> samples. Focusing testing <strong>on</strong> this age group<br />

could also save resources and maximize efficiency in testing and culling programmes.<br />

INTRODUCTION<br />

It is generally accepted that paratuberculosis (PTB) is a slow infecti<strong>on</strong> that begins during <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

first weeks <str<strong>on</strong>g>of</str<strong>on</strong>g> life in a c<strong>on</strong>taminated envir<strong>on</strong>ment, but that do not develop into visible lesi<strong>on</strong>s<br />

and disease until <str<strong>on</strong>g>the</str<strong>on</strong>g> early adult life <str<strong>on</strong>g>of</str<strong>on</strong>g> cows in some animals.<br />

Sensitivity <str<strong>on</strong>g>of</str<strong>on</strong>g> PTB diagnosis varies with <str<strong>on</strong>g>the</str<strong>on</strong>g> stage <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> infecti<strong>on</strong> and lack <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

sensitivity <str<strong>on</strong>g>of</str<strong>on</strong>g> diagnostic tests is <str<strong>on</strong>g>the</str<strong>on</strong>g> main hurdle for <str<strong>on</strong>g>the</str<strong>on</strong>g> successful use <str<strong>on</strong>g>of</str<strong>on</strong>g> test and cull<br />

strategies.<br />

In this slaughterhouse study, <str<strong>on</strong>g>the</str<strong>on</strong>g> main goal was to determine <str<strong>on</strong>g>the</str<strong>on</strong>g> age structure <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

Mycobacterium avium subsp. paratuberculosis (MAP) infecti<strong>on</strong> in Friesian cattle as well as to<br />

obtain new data <strong>on</strong> <str<strong>on</strong>g>the</str<strong>on</strong>g> efficacy <str<strong>on</strong>g>of</str<strong>on</strong>g> four diagnostic tests for detecti<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> subclinically infected<br />

animals.<br />

MATERIALS AND METHODS<br />

Animal selecti<strong>on</strong> and sampling<br />

Using a weekly systematic sampling, 581 Friesian animals were examined from March 2007<br />

to December 2008, at two local slaughterhouses in <str<strong>on</strong>g>the</str<strong>on</strong>g> Basque Country. After stunning,<br />

blood samples from <str<strong>on</strong>g>the</str<strong>on</strong>g> jugular vein were collected into sterile tubes c<strong>on</strong>taining EDTA.<br />

Additi<strong>on</strong>ally, fresh and formalin-fixed samples <str<strong>on</strong>g>of</str<strong>on</strong>g> jejunal caudal lymph node (JCL) and distal<br />

ile<strong>on</strong> and ileocecal valve (ICV) were aseptically taken. Bovine identificati<strong>on</strong> documents (BID)<br />

were reviewed for age data collecti<strong>on</strong>.<br />

ELISA<br />

The Pourquier® ELISA <strong>Paratuberculosis</strong> Antibody Screening kit (Institut Pourquier,<br />

M<strong>on</strong>tpellier, France) was used <strong>on</strong> plasma samples according to <str<strong>on</strong>g>the</str<strong>on</strong>g> manufacturer’s<br />

instructi<strong>on</strong>s. Samples with positive and doubtful results for <str<strong>on</strong>g>the</str<strong>on</strong>g> screening ELISA were<br />

retested using Pourquier® ELISA <strong>Paratuberculosis</strong> Antibody Verificati<strong>on</strong> kit (Institut<br />

Pourquier, M<strong>on</strong>tpellier, France). Positive results corresp<strong>on</strong>ded to sample to positive (S/P)<br />

ratios above 70%.<br />
