-possible link <str<strong>on</strong>g>of</str<strong>on</strong>g> MAP infecti<strong>on</strong> with Crohn’s disease<br />

-development <str<strong>on</strong>g>of</str<strong>on</strong>g> new or optimizati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> existing experimental infecti<strong>on</strong> models<br />

-discovery <str<strong>on</strong>g>of</str<strong>on</strong>g> new antigens<br />

-development <str<strong>on</strong>g>of</str<strong>on</strong>g> new vaccines based <strong>on</strong> live attenuated whole bacteria or <strong>on</strong> sub-unit comp<strong>on</strong>ents (protein and<br />

DNA)<br />

The interacti<strong>on</strong> between <str<strong>on</strong>g>the</str<strong>on</strong>g> early, innate immune resp<strong>on</strong>se and <str<strong>on</strong>g>the</str<strong>on</strong>g> cognate acquired immune resp<strong>on</strong>se had not<br />

been studied in much detail so far for infecti<strong>on</strong>s with M. avium subsp paratuberculosis. More and more results<br />

now suggest that a malfuncti<strong>on</strong>ing innate resp<strong>on</strong>se, caused by an immature system in young animals and/or by<br />

genetic factors (a.o. deficient reactivity to mycobacterial peptidoglycan as observed in Crohn’s patients) would<br />

lie at <str<strong>on</strong>g>the</str<strong>on</strong>g> basis <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> deficient cell mediated immune (CMI) resp<strong>on</strong>se observed in animals with multibacillary<br />

forms <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> disease. The evidence for a role <str<strong>on</strong>g>of</str<strong>on</strong>g> Th2 and regulatory T cells in <str<strong>on</strong>g>the</str<strong>on</strong>g> decreased CMI resp<strong>on</strong>ses, so<br />

characteristic <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>the</str<strong>on</strong>g> clinical phase <str<strong>on</strong>g>of</str<strong>on</strong>g> Johne’s disease, is questi<strong>on</strong>ed and although high levels <str<strong>on</strong>g>of</str<strong>on</strong>g> IL-10 can be<br />

detected, it are CD14+ MHC class II+ m<strong>on</strong>ocytes ra<str<strong>on</strong>g>the</str<strong>on</strong>g>r than T cells that produce this cytokine and moreover,<br />

even at early stages <str<strong>on</strong>g>of</str<strong>on</strong>g> infecti<strong>on</strong> IL-10. On <str<strong>on</strong>g>the</str<strong>on</strong>g> o<str<strong>on</strong>g>the</str<strong>on</strong>g>r hand, <str<strong>on</strong>g>the</str<strong>on</strong>g> role <str<strong>on</strong>g>of</str<strong>on</strong>g> ano<str<strong>on</strong>g>the</str<strong>on</strong>g>r Th subset, i.e. Th17 may be more<br />

relevant, as gene expressi<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> interleukin17 and interleukin 23 was found to be upregulated in an experimental<br />

infecti<strong>on</strong> model <str<strong>on</strong>g>of</str<strong>on</strong>g> red deer during disease progressi<strong>on</strong>. This Th17 populati<strong>on</strong> has also been incriminated in<br />

Crohn’s disease and o<str<strong>on</strong>g>the</str<strong>on</strong>g>r auto-immune diseases.<br />

With respect to vaccinati<strong>on</strong>, it is clear that live attenuated vaccines can be very effective, but a balance has to be<br />

found between <str<strong>on</strong>g>the</str<strong>on</strong>g>ir immunogenicity (less attenuated strains may be more immunogenic but may interfere with<br />

<str<strong>on</strong>g>the</str<strong>on</strong>g> immunodiagnostic tests) and <str<strong>on</strong>g>the</str<strong>on</strong>g>ir vaccine potential. Even for subunit vaccines <str<strong>on</strong>g>the</str<strong>on</strong>g> interference with existing<br />

diagnostics may be an issue. Characterizati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> immunodominant and specific antigens <str<strong>on</strong>g>of</str<strong>on</strong>g> MAP will be essential<br />

to overcome this hurdle.<br />
