“weight-of-evidence.”.In.<strong>the</strong>.case.of.bisphenol.<br />

A,.evidence.from.<strong>the</strong>.limited.number.of.studies.in.humans.exposed.to.bisphenol.A.is.not.sufficient.to.reach.c<strong>on</strong>clusi<strong>on</strong>s.regarding.possible.developmental.or.reproductive.hazard..In.<br />

c<strong>on</strong>trast,.<strong>the</strong>re.is.a.large.literature.of.laboratory.<br />

animal.studies..These.include.studies.of.traditi<strong>on</strong>al.designs.carried.out.to.assess.<strong>the</strong>.toxicity.of.bisphenol.A,.as.well.as.a.wide.variety.of.studies.examining.<strong>the</strong>.possibility.that.exposure.to.<br />

Many.of.<strong>the</strong>.laboratory.animal.studies.of.bisphenol.A.have.technical.or.design.shortcomings.or.<strong>the</strong>ir.reports.do.not.provide.sufficient.experimental.details.to.permit.an.assessment.of.technical.adequacy.(2)..As.discussed.in.more.detail.<br />

Ra<strong>the</strong>r,.in.an.effort.to.glean.informati<strong>on</strong>.that.<br />

reported.effects.of.bisphenol.A,.NTP.evaluated.<br />

many.individual.study.reports..Attenti<strong>on</strong>.was.<br />

paid.to.issues.of.sample.size,.c<strong>on</strong>trol.for.litter.<br />

Historically,.bisphenol.A.has.been.characterized.<br />

most. comm<strong>on</strong>. type. of. positive. c<strong>on</strong>trol. compounds.used.in.bisphenol.A.studies.are.potent.<br />

estrogens..There.is.wide.variability.in.in vitro<br />

estrogenic.potency.estimates.for.bisphenol.A,.<br />

although.<strong>the</strong>.mean.estimate.is.~1,000.to.10,000.<br />

times. less. potent. than. positive. c<strong>on</strong>trol. compounds.(2)..However,.a.number.of.<strong>the</strong>.“low”.<br />

dose. studies. suggest. that. bisphenol.A. has. a.<br />

higher.in vivo.potency.than.would.be.predicted.<br />

based.<strong>on</strong>.binding.to.estrogen.receptor.alpha..<br />

The.lack.of.c<strong>on</strong>cordance.in.potency.estimates.<br />

based.<strong>on</strong>.estrogen.receptor.binding.<strong>and</strong>.in vivo.<br />

biological.activity.has.been.a.point.of.debate.
