1.12 Estimation of protein
1.12.1 Alkaline copper reagent
50 mL of reagent A and I mL of reagent B were mixed fresh at the time of
use.
Reagent A
2 g of sodium carbonate was dissolved in and made up to 100 mL with 0.1 N
sodium hydroxide solution.
Reagent B
5 mg of copper sulfate in 1 mL of 4% sodium potassium tartarate.
1 .I 2.2 Sodium hydroxide solution (0.1 N)
0.4 g of sodium hydroxide was dissolved in and made up to 100 mL with
distilled water.
1.12.3 Sodium potassium tamirate (4%)
4 g sodium potassium tartarate was dissolved in and made up to 100 mL with
distilled water.
1.12.4 Bovine serum albumin (BSA) standard
100 mg of bovine serum albumin was dissolved in and made up to 100 mL
with distilled water. This solution was diluted 10 times to obtain a
concentration of 0.1 mg/ mL.
1.12.5 Folin Ciocalteau reagent (IN)
Commercial Folin Ciocalteau reagent was diluted with equal volume of
distilled water.
