They bind to the gastric proton pump on the parietal cellmembrane, inhibiting the release of hydrogen ions from the parietal cells into the lumenof the gastric glands and hence stomach. 44 Some of the adverse effects of PPIs includenausea, diarrhea, dizziness 45 , hypergastrinemia 46 , enteric infections 2 etc. It has beendemonstrated that irreversible inhibition of H + /K + -ATPase occurs following acidactivation of these compounds within the acidic compartments in the parietal cells andcovalent binding of the reactive intermediate to one or more critical thiol groups on theenzymes present in apical membrane 47 as in Figure 9. Acid secretion is therefore blockedat the final step of its production independent of the different kind of its stimulation. 4814
