876 CONCLUSÕESOs estudos <strong>de</strong> mo<strong>de</strong>lagem comparativa foram realizados <strong>de</strong> forma a obter a <strong>estrutura</strong>tridimensional da protease do HSV-1, cuja <strong>estrutura</strong> não se encontra elucidada na literatura. Omo<strong>de</strong>lo foi obtido e validado para ser utilizado na simulação <strong>de</strong> <strong>docking</strong> molecular.Os resultados <strong>de</strong> <strong>docking</strong> molecular mostraram cor<strong>relação</strong> com os valores <strong>de</strong> ativida<strong>de</strong>dos compostos, confirmando a importância, <strong>de</strong> alguns resíduos para a ativida<strong>de</strong> da protease. Oresíduo Ser129 parece ser o mais importante da tría<strong>de</strong> catalítica, seguido dos resíduos His61,His148, Arg157 e Arg156, uma vez que o composto mais ativo (26) foi o único da série <strong>de</strong>benzoxazinonas que apresentou interação com o resíduo Ser129 e outros compostos com altaativida<strong>de</strong> apresentaram interações com estes outros resíduos. Interações hidrofóbicas, nãoobservadas até o momento na literatura, entre uma cavida<strong>de</strong> hidrofóbica composta pelosresíduos Leu27, Ala28, Leu38, Ala39, Val28, Leu130, Ala131, Ala150, Leu151, Ile154,Gly155, Leu158 e Gly159 e as benzoxazinonas avaliadas sugerem a importância <strong>de</strong>ssacavida<strong>de</strong> para a ativida<strong>de</strong> da protease <strong>de</strong> HSV-1.Os estudos <strong>de</strong> Druglikeness e Drugscore realizados no programa Osiris indicaram ocomposto 29 para a realização <strong>de</strong> modificações <strong>estrutura</strong>is para obtenção <strong>de</strong> um protótipo.Apesar disto, o composto 26 foi também consi<strong>de</strong>rado <strong>de</strong>vido à sua alta ativida<strong>de</strong>.Foram propostos quatro compostos por técnicas <strong>de</strong> hibridização molecular ebioisosterismo que foram analisados através <strong>de</strong> um estudo <strong>de</strong> <strong>docking</strong> molecular. Esse estudoapontou a molécula 34 como protótipo, indicando um possível aumento da ativida<strong>de</strong> em<strong>relação</strong> aos compostos 26 e 29. Apesar <strong>de</strong>sta evidência, os estudos <strong>de</strong> Druglikeness eDrugscore dos compostos propostos mostraram resultados inferiores aos do composto 29.Desta forma, outros ensaios experimentais <strong>de</strong>vem ser realizados para sustentar a hipótese doaumento da ativida<strong>de</strong> do composto 34 em <strong>relação</strong> às <strong>de</strong>mais benzoxazinonas.
