Cytotoxic drugs
- Cytotoxic drugs all kill the cells that are capable of self replication e.g lymphocytes.
- They can be classified according to stage of action as :
Phase specific ( i.e toxic for cells entering the particular phase of cell cycle) e.g
o Azathioprine & methotrexate toxic for cells in S (DNA synthesis) phase
Cycle specific : (toxic for cells at all stages of cell mitotic cycle) e.g
o Cyclophosphamide & chlorambucil.
- They can be classified according to mechanism of action as :
A. Alkylating agents
Cytotoxic Mechanism of action Adverse effects
Cyclophosphamide
(Endoxan)
- Alkylating agent - causes
Cross-linking in DNA
1) Haemorrhagic cystitis (can be by
2) Myelosuppression
3) Transitional cell carcinoma
4) Gonadal hypoplasia & infertility.
B. Cytotoxic antibiotics
Cytotoxic Mechanism of action Adverse effects
Bleomycin - Degrades preformed DNA Pulmonary fibrosis
Doxorubicin
(Adriamycin)
- Stabilizes DNA-topoisomerase II complex
inhibits DNA & RNA synthesis
