Procedure

1. Wash hands and don gloves.

2. Assemble necessary equipment and supplies.

3. Using a serological pipette, place into cuvettes the following volumes (mL) and

mix well. The volumes may be increased if the instrument requires volumes

greater than 1.0 mL.

4. Incubate all cuvettes at 37°C for 10 minutes, or incubate at room temperature for

20 minutes.

5. Read S (standard) and RB (reagent blank) at 500 nm within 15 minutes.

6. Discard all supplies used for the procedure in the appropriate containers. A disinfectant

should be used to clean the work surfaces and equipment should be

cleaned and restored to its former position. Gloves should be discarded appropriately

and the hands washed thoroughly in accordance with established policies.

Quality Control

Two levels of control material, including both a normal and an abnormal value, with

known glucose levels, should be analyzed by this method on each day of testing.

Reporting of Results

Results are obtained using the following equation:

Calculation

Glucose (mg/dL) = Absorbance of unknown (Au) 100

Absorbance of standard (As)

where Au is the absorbance value of the unknown, As is the absorbance value of

the standard, and 100 is the concentration of the standard in milligrams per deciliter

(mg/dL).

Results are reported in milligrams per deciliter (mg/dL). In some countries, they are

reported in SI (International System) units that are derived from the metric system.

Conversion to SI units is often performed by the instrument upon which the test is

performed. Some health care facilities report values in both SI and metric units of

measurement.
