Equipment and Supplies

1. Gloves, disposable paper towels, and disinfectant or other cleaning solution

2. Report form provided by your instructor

3. Serum (preferred because fibrinogen is contained in a plasma sample)

a. For accuracy, the sample must be free of significant hemolysis.

b. A serum sample that is free from gross hemolysis or lipemia will remain

stable for up to one week at room temperature and for 30 days at 2°C–8°C.

c. When a blood sample is centrifuged immediately after clotting and the

serum is removed from the blood cells, hemolysis is greatly reduced.

4 Micropipetter with disposable pipette tips for measuring patient specimen as μLs

5. Serological pipettes

6. Stanbio kit for total protein (contains the reagent copper sulfate pentahydrate in

aqueous sodium hydroxide and aqueous protein standard [l0 mg/dL])

7. Spectrophotometer capable of absorbance readings at 550 nm (Figure 14-2)

8. Water bath or heat block set at 37°C (optional) (The reaction takes 10 minutes at

room temperature, and approximately 3 minutes at 37°C.)

9. Cuvettes

10. 13 × 100 test tubes and test tube rack

11. Waterproof sharp marker for labeling test tubes

12. Vortex mixer (optional) (desirable since the complex of the dye and proteins will

fall to the bottom of the tube in which the specimen and reagents are mixed and

even in the cuvettes before reading them on the spectrophotometer)

13. Electronic timing device or clock/watch with second hand

Procedure

1. Wash hands and don gloves.

2. Assemble necessary equipment and supplies.
