Equipment and Supplies

1. Gloves, disposable paper towels, and disinfectant

or other cleaning solution

2. Report form

3. EDTA anticoagulated blood sample or a

freshly clotted sample

4. Waterproof sharp marker

5. Normal physiological (0.85%) saline (blood

bank quality) in squeeze bottle

6. Disposable Pasteur pipettes or transfer

pipettes

7. Anti-A blood serum

9. Applicator sticks or plastic stirrers

10. Clean microscope slides or specialized cell typing slides

11. Illuminating lamp with magnifying mirror (optional)

12. Electronic timing device or clock/watch with second hand

13. Disposal containers appropriate for biohazardous waste and sharps container

Procedure

1. Wash hands and put on gloves.

2. Assemble equipment and materials.

3. Mark a clean slide into two halves, labeling the left side “A” and the right

side “B.”

4. Using a Pasteur pipette, transfer pipette, or dropper included with the reagent, place

one drop of anti-A serum on the “A” side (do not allow the dropper to touch slide).

5. Using a Pasteur pipette, transfer pipette, or dropper included with the reagent, place

one drop of anti-B serum on the “B” side (do not allow the dropper to touch slide).

8. Add one drop of well-mixed blood to each side of the slide using the Pasteur

pipette or a pair of applicator sticks (the drop of blood should be no larger than

the drop of antiserum).

9. Using a clean applicator stick, mix the blood and antiserum on side A and spread

into a smooth round circle about the size of a nickel.

10. Using a clean applicator stick, repeat the procedure on side B.

11. Rock the slide gently for 2 minutes and look for agglutination. An illuminating

lamp with magnifying mirror may be used as an optional practice.

12. Record which sides of the slide show agglutination, using the following form.

Place a plus sign (“+”) for agglutination into the appropriate blank(s) and a minus

sign (“–“) to indicate no agglutination.

13. Discard all supplies used for the procedure in the appropriate containers.
