18. Clean the microscope stage with lens paper or tissue. Clean the condenser if

necessary.

19. Retain the slide if required.

20. Discard all supplies used for the procedure in the appropriate containers. A disinfectant

should be used to clean the work surfaces and equipment should be

cleaned and restored to its former position. Gloves should be discarded appropriately

and the hands washed thoroughly in accordance with established policies.

Reporting of Results

Calculate the percentage of each of the five basic leukocytes (neutrophils, eosinophils,

basophils, lymphocytes, and monocytes). Report the direct leukocyte count

(DLC) as a percentage. If the total WBC is known, the count may also be reported in

absolute numbers by multiplying the percentages of each type by the actual WBC

count. Under “Comments”, note the presence of any immature cells, especially blast

cells (early immature types), and report these as percentages also.

In addition, the numbers and morphology of platelets can also be evaluated along

with the RBCs. Report your findings from the Practical White Blood Cell Differentiation

procedure in the Procedure #2 Report Form or other form supplied by your instructor.

Evaluation of Student Results for Procedure #2

The differentials have been evaluated by counting 200 cells. Grades will be determined

as follows:

1. Segmented neutrophils and lymphocytes must come within ±2% of the target

value determined by the instructor. In the case that the total percentage is

extremely low (i.e., a lymphocyte count of less than 10%), two cells on either side

of the target value will be acceptable.

2. One letter grade will be deducted for EACH type of cell that is not within an

acceptable range.
