A disinfectant should be used to clean the work surfaces and equipment should

be cleaned and restored to its former position. Gloves should be discarded

appropriately and the hands washed thoroughly in accordance with established

policies.

Reporting of Results

Interpret your results for ABO direct blood grouping according to the following

criteria.

Agglutination in Tube A = Group A

Agglutination in Tube B = Group B

Agglutination in Tubes A and B = Group AB

No agglutination in Tubes A or B = Group O

Direct Blood or Forward Grouping by Slide Method

Principles

The quality and care practiced by the laboratory professional is critical in the blood

bank, as a mistake has an immediate impact upon the patient in many instances.

These mistakes can be of misidentification, misinterpretation, or in the performance

of the testing. Slide tests, like tube tests for ABO grouping, employ the same technique

of introducing antigens on the surfaces of RBCs to anti-sera that contain

antibodies that correspond to the antigens for a specific blood group. If the antigen

that correlates with the antibody against that particular antigen is not present on the

RBCs, no reaction occurs.

The direct grouping performed on a microscope slide is the quickest and easiest

method to determine the ABO grouping (Figure 17-2) but is not the choice for most

blood banking professionals. The problem with this procedure lies in the possibility

of proteins interfering with the reaction, which is avoided in the tube method, as the

RBCs that contain the A or B antigen(s) are washed before testing.
