Body<br />

Objectives: The aim of this study w<strong>as</strong> to examine synostosis, cranial b<strong>as</strong>e- length and skull<br />

<strong>as</strong>ymmetry in PLCγ2 knock-out mice <strong>as</strong> a model for osteopetrosis.<br />

Methods: In this study, four 135-days-old and one 24-days-old Phospholypace C gamma 2<br />

(PLCγ2) knockout mice – and wild-type mice for comparison– were examined. The newborn<br />

mice were tested by PCR, then Western blot h<strong>as</strong> showed PLCγ2 expression. The length of the<br />

cranial b<strong>as</strong>e and the <strong>as</strong>ymmetry of the skull were me<strong>as</strong>ured by MCT (60 kV, 0,5° rotation step,<br />

14 µm voxel size). Hystomorphometric analysis w<strong>as</strong> used to demonstrate the developmental<br />

stage of the synostosis while double staining with Alcian Blue and Alizarin Red S indicated the<br />

synchondrosis and the ossification centers, respectively. Morphologic parameters were determined<br />

from 3D rendered images using DataViewer (Skyscan) software.<br />
