2. Incubate the root cultures in the dark <strong>and</strong> the epicotyl cultures either in the<br />

light or in the dark.<br />

3. After 45 days the globular calli are collected to determine the fresh <strong>and</strong> dry<br />

mass.<br />

High callus fresh weight is obtained from root segments cultured on 1.15–2.3 µM 2,<br />

4-D. The epicotyl segments cultured in the light produce superior values <strong>of</strong> callus<br />

fresh weight at all concentrations <strong>of</strong> 2,4-D or at 2.7 µM NAA; in the dark the best<br />

concentration <strong>for</strong> callus induction is either 4.6 µM 2,4-D or 2.7 µM NAA.<br />
