Culture medium. The shoot proliferation culture medium, supplemented with 3%<br />

sucrose <strong>and</strong> solidified with 0.6% agar, is also used <strong>for</strong> the in vitro introduction <strong>and</strong><br />

establishment <strong>of</strong> axillary shoots (Hispanlab, S.A.). The PGRs used in the<br />

introduction medium are 1.76 µM BA <strong>and</strong> 0.2 µM indole-3-butyric acid (IBA). The<br />

medium is dispensed in 25 × 150 mm culture tubes covered with permeable membrane<br />

caps. Sterilization <strong>of</strong> media <strong>and</strong> culture conditions are described above. Axillary<br />

buds sprout in this medium (Figure 3C, D) <strong>and</strong>, after removing contaminated nodes,<br />

elongated shoots are transferred to jars containing proliferation medium (Figure 4).<br />
