MICROGRAFTING OF PISTACHIO<br />

6. Incubate the cultures in a growth room at 40 µmol m –2 s –1 photosynthetic<br />

photon flux intensity (fluorescent lamps, 75W) <strong>and</strong> a photoperiod <strong>of</strong> 16 h at<br />

25 ± 2°C <strong>for</strong> 10–14 days. Use the seedlings (Figure 1B) as rootstocks <strong>for</strong><br />

grafting.<br />

2.2.4. Development <strong>of</strong> Grafting Method<br />

Various grafting procedures were tried within the culture vessels without removing<br />

the rootstocks from the medium (Figure 2):<br />

1. Slit micrografting: The rootstock was decapitated to remove all leaves <strong>and</strong><br />

a vertical slit was made on the stump, <strong>and</strong> the scion base, cut in v-shape,<br />

was fitted in the slit.<br />

2. Wedge micrografting on the stump: The rootstock was decapitated to remove<br />

all leaves. A wedge was cut in the stump, <strong>and</strong> the scion base cut in a v<br />

shape, was gently fit into the wedge.<br />

3. Wedge micrografting in the leaf axil: The rootstock was decapitated leaving<br />

a single leaf. A wedge was cut <strong>of</strong>f in the leaf axil after removing the leaf<br />

blade <strong>and</strong> the scion base gently sloped to fit into the wedge.<br />

similar to the seed germination medium. In the subsequent experiments, slit micrografting<br />

was elected <strong>for</strong> grafting. In this method, the rootstock was decapitated to<br />

remove all leaves <strong>and</strong> a vertical slit was made on the stump, <strong>and</strong> the scion base, cut<br />

in v-shape, was inserted in the slit (Figure 1C).<br />

Different scion sizes were tested: 10 mm long.<br />

There were differences in the frequencies <strong>of</strong> successful micrografts between the<br />

treatments applied (Onay et al., 2004b). Success increased with the size <strong>of</strong> the scions<br />

up to 6 mm but then decreased with size. All shoot tips
