2.8. Pretreatment <strong>and</strong> Encapsulation Matrix<br />

Prior to encapsulation the shoots should be soaked in liquid White’s (1954) basal<br />

medium supplemented with sucrose (1%), NAA (10.8 µM ) <strong>and</strong> IBA (39.4 µM) <strong>and</strong><br />

agitated at 100 rpm (on a gyratory shaker) <strong>for</strong> 48 h. The shoots can be blot dried on<br />

a filter paper, mixed in a gel <strong>of</strong> 3% sodium alginate (Sigma) prepared in MS basal<br />

medium or SMM <strong>and</strong> encapsulated using CaCl2·2H2O (1.36g / 150 ml) solution.
