2.4. Incubation <strong>of</strong> Cultures <strong>and</strong> Maintenance<br />

Maintain cultures in the culture room at 25 ± 2°C with 55 ± 5% relative humidity.<br />

Programme <strong>for</strong> 16 h photoperiod, emitted by 40 W Philips tubes. Transfer explants<br />

to a freshly prepared (same) culture medium after every 8 weeks. In some cases,<br />

frequent subculturing may be helpful. Allow explants to grow <strong>for</strong> a longer duration,<br />

even up to 16–18 weeks, without subculturing, if the medium does not dry up.<br />
