CARCINOGENIC EFFECTS<br />

Human Studies<br />

No data are available addressing the carcinogenicity of 2,4-dinitrotoluene (2,4-DNT) in humans.<br />

Animal Studies<br />

Lee et al. (1978) exposed male and female CD rats to practical grade 2,4-DNT in feed at<br />

concentrations of 0, 15, 100, and 700 ppm for 720 days. Animals were sacrificed at 750 days.<br />

Decreased weight gain and lifespan were observed among animals in the highest treatment group.<br />

Noncancer toxic effects observed included toxic anemia and aspermatogenesis. Tumors observed<br />

included fibromas of the connective tissue of male rats and fibroadenoma of the mammary gland of<br />

female rats. The incidence data for liver and mammary tumors in female rats are presented in Table 1.<br />

Significantly increased incidence was found in the highest dose group for liver tumors (p = 4 × 10 -4 ;<br />

Fisher’s exact test), mammary gland tumors (p = 1.75 × 10 -4 ), and combined mammary gland and<br />

liver tumors (p = 7 × 10 -5 ). Mammary tumor incidence among male rats was 1/37, 0/37, 0/29, and<br />

17/23 in the 0, 15, 100, and 700 ppm 2,4-DNT dose groups, respectively. Only the highest dose<br />

group showed significantly increased tumor incidence (p = 4 × 10 -9 ).<br />
