IV. DERIVATION OF CANCER POTENCY Basis for Cancer Potency The US EPA (1980) derived a cancer potency value based on the tumor incidence data in the study by Lee et al. (1978) showing the induction of liver and mammary tumors in female CD rats. This study was selected over the NCI (1978) study because of published reservations by NCI concerning the adequacy of the study for estimating cancer potency in humans. Methodology The US EPA (1980) calculated a “transformed” dose rate of 0, 0.71, 3.9, and 34.0 mg/kg-day for the animals in the study by Lee et al. (1978) exposed to 0, 15 100 and 700 ppm 2,4-DNT in their diet, respectively. A linearized multistage procedure was applied to the combined mammary gland and liver tumor incidence data presented in Table 1 in order to calculate an animal cancer potency value (q animal ). The calculated q animal was 0.058 (mg/kg-day) -1 . The q animal was converted to a human cancer potency (q humna ) based on the following relationship, where bw animal is the assumed body weight for the test species (Lee et al., 1978; bw animal = 0.464 kg) and bw human is the assumed human body weight (70 kg): The resulting q human is 0.31 (mg/kg-day) -1 . q human = q animal × (bw h /bw a ) 1/3 A unit risk value based upon air concentrations was derived by <strong>OEHHA</strong>/ATES using an assumed human breathing rate of 20 m 3 /day, 70 kg human body weight, and 100% fractional absorption after inhalation exposure.
