• Toxic effect <strong>on</strong> terrestrial organisms: acute toxicity in worms, effect<br />

<strong>on</strong> soil respirati<strong>on</strong>, effect <strong>on</strong> amm<strong>on</strong>ificati<strong>on</strong>, effect <strong>on</strong> nitrificati<strong>on</strong>,<br />

effect <strong>on</strong> asymbiotic N-fixati<strong>on</strong> <strong>and</strong>, in some cases, effect <strong>on</strong><br />

symbiotic N-fixati<strong>on</strong>.<br />

• Toxic effect <strong>on</strong> birds: toxicity caused by food intake in species with<br />

different food resources (2 species), reproducti<strong>on</strong> test (1 species).<br />

The following toxicological data for assessment of effects <strong>on</strong> humans<br />

must be provided for <str<strong>on</strong>g>the</str<strong>on</strong>g> active ingredient:<br />

1) Acute toxicity<br />

Toxicity with a single dose of <str<strong>on</strong>g>the</str<strong>on</strong>g> substance. Measured as LD50 (<str<strong>on</strong>g>the</str<strong>on</strong>g> dose<br />

needed for half <str<strong>on</strong>g>the</str<strong>on</strong>g> animals to die). Must be tested in separate tests with<br />

swallowing, skin c<strong>on</strong>tact <strong>and</strong> inhalati<strong>on</strong>.<br />

2) Local irritati<strong>on</strong><br />

Skin irritati<strong>on</strong>, eye irritati<strong>on</strong> <strong>and</strong> allergenic properties in c<strong>on</strong>necti<strong>on</strong> with<br />

skin c<strong>on</strong>tact must also be investigated.<br />

3) <str<strong>on</strong>g>Sub</str<strong>on</strong>g>-chr<strong>on</strong>ic toxicity<br />

Toxicity with daily dosing for 3-6 m<strong>on</strong>ths. Two tests are required: <strong>on</strong>e <strong>on</strong><br />

rats (of 3 m<strong>on</strong>ths durati<strong>on</strong> ) <strong>and</strong> <strong>on</strong>e <strong>on</strong> n<strong>on</strong>-rodents (e.g. dogs) of 3-6<br />

m<strong>on</strong>ths durati<strong>on</strong>.<br />

4) Chr<strong>on</strong>ic toxicity<br />

Toxicity with l<strong>on</strong>g-term dosing. Chr<strong>on</strong>ic toxicity <strong>and</strong> carcinogenicity are<br />

often combined in <str<strong>on</strong>g>the</str<strong>on</strong>g> same 2-year test. Two tests are required <strong>on</strong><br />

different species of mammal. If dogs are more sensitive than rats, a<br />

separate 1-year toxicity test <strong>on</strong> dogs is required.<br />

5) Carcinogenic effect<br />

6) Mutagenicity<br />

The ability of <str<strong>on</strong>g>the</str<strong>on</strong>g> substance to cause damage to <str<strong>on</strong>g>the</str<strong>on</strong>g> genetic material.<br />

Must be tested in test-tube tests (in vitro) <strong>and</strong> in animal tests (in vivo). If<br />

<str<strong>on</strong>g>the</str<strong>on</strong>g> substance is found to be positive in <str<strong>on</strong>g>the</str<strong>on</strong>g> above tests, it must be tested<br />

in gametes.<br />

7) Tests <strong>on</strong> impairment of fertility<br />

Multi-generati<strong>on</strong>al tests; <str<strong>on</strong>g>the</str<strong>on</strong>g> possible ability of <str<strong>on</strong>g>the</str<strong>on</strong>g> substance to reduce<br />

fertility must be tested by feeding with <str<strong>on</strong>g>the</str<strong>on</strong>g> substance through several<br />

generati<strong>on</strong>s.<br />

8) Teratogenicity tests<br />

The ability of <str<strong>on</strong>g>the</str<strong>on</strong>g> substance to damage <str<strong>on</strong>g>the</str<strong>on</strong>g> foetus during pregnancy must<br />

be tested in two species of mammal. The pregnant dam must receive <str<strong>on</strong>g>the</str<strong>on</strong>g><br />

substance by tube in <str<strong>on</strong>g>the</str<strong>on</strong>g> most sensitive period of <str<strong>on</strong>g>the</str<strong>on</strong>g> pregnancy.<br />

9) Neurotoxicity<br />

Particularly for <str<strong>on</strong>g>the</str<strong>on</strong>g> so-called cholinesterase inhibitors.<br />

10) Toxicity of any metabolites, degradati<strong>on</strong> products <strong>and</strong> impurities.<br />
