Figure 3. Whole-cell hybridization of fixed cells from 3 samples (T2, T7 and T28) collected<br />

from the enrichment culture in bioreactor. Cells were stained with DAPI (T2A, T7A and T28<br />

A), and hybridized with the FITC-labeled Eub338 probe (T7B), or the Cy-3-labeled Arch 915<br />

(T2B and T28B). Cells were viewed by epifluorescence microscopy in which DAPI-, FITCand<br />

Cy-3-specific filters were used. The relative proportions of archaeal and bacterial cells<br />

were <strong>de</strong>termined by counting approximately 2000 cells on filter and resulted in: 99.0% of the<br />

cells <strong>de</strong>tected at T2 belonged to the Archaea (see T2B), 99.0% of the cells <strong>de</strong>tected at T7<br />

belonged to the Bacteria (see T7B), 40% of the cells <strong>de</strong>tected at T28 belonged to the<br />

Archaea (see T28B). Scale bars = 10 µm.<br />
