DISKUSSION 115 Zunahme der Glutamat-Decarboxylase GadB (von 0,3 auf 1,9) bei gleichzeitiger Abnahme der Glutamat-Decarboxylase GadA (von 5,5 auf 2,1) . Dies deutet auf eine unterschiedliche Regulation von gadA und gadB sowie eventuell auch auf unterschiedliche Eigenschaften der Enzyme hin . Auffällig war bei den Proteomanalysen der Fed-Batch-Fermentationen, dass mehrere Proteine, die in einer phosphorylierten und einer dephosphorylierten Form vorliegen können, veränderte Intensitäten zeigten (Icd, DnaK, YfiD, GroL, TufA, UspA) . Die Proteinphosphorylierung ist ein wichtiger Mechanismus für die Kontrolle von Protein- Aktivitäten . In Eukaryonten gibt es Proteinkinasen, die <strong>durch</strong> verschiedenste Umweltstresse aktiviert werden und eine Adaptation an diese Stressbedingungen ermöglichen (Tamura et al., 2002 ; Tibbles & Woodgett, 1999) . Das universelle Stressprotein UspA von E. coli (Freestone et al ., 1997) wird unter anderem dann vermehrt gebildet, wenn die Zelle toxischen Substanzen wie Schwermetallen, Säuren oder Antibiotika ausgesetzt ist (Nystrom & Neidhardt, 1992) . Die Zunahme der UspA-Konzentration im Verlaufe der al-Fermentation kann also wiederum als Folge des Säurestresses betrachtet werden . In Zellen der al-Fermentation war UspA schon zum Zeitpunkt der ersten Probennahme in relativ hoher Intensität nachweisbar und nahm im Verlauf der Fermentation noch weiter zu . Das DNA-bindende Protein H-NS beeinflusst die Expression vieler Gene . Es wird postuliert, dass es eine Rolle bei der Virulenz, beim Kälteschock, beim pH-Stress und der Osmoregulation spielt (Higgins et al., 1990) . Die Zunahme von H-NS in der al-Kultur könnte die negative Autoregulation widerspiegeln, die in der logarithmischen Wachstumsphase, nicht aber in der stationären Phase beobachtet wurde (Falconi et al ., 1993) .
