Symptom Cause Solution<br />

Pipetting problems Wells inoculated unevenly Dispense first aliquot from pipettor back into reservoir.<br />

Depress lever smoothly and completely.<br />

Make sure tips are lined up evenly and tightly seated.<br />

Avoid pipetting or trapping air bubbles.<br />

Visually check wells after inoculating.<br />

Incorrect volumes used<br />

Incubating MicroPlates<br />

100 µl for anaerobic bacteria, yeasts and fungal.<br />

150 µl for aerobic bacteria.<br />

Symptom Cause Solution<br />

Poor growth or poor<br />

pattern formation in<br />

MicroPlates<br />

Wells at the four<br />

corners or edges of<br />

the MicroPlate have<br />

lower liquid levels<br />

after incubation<br />

Wrong incubation<br />

incubated in a closed<br />

Incubate according to specified temperature and<br />

conditions. See Section 4 and Appendices.<br />

Incubate the MicroPlate at the same temperature as<br />

the growth plate.<br />

Make sure incubator humidity is sufficient.<br />

Add a pan of water to humidify incubator, or incubate<br />

MicroPlates in a plastic container humidified with<br />

moist paper towels.<br />

Make sure you have the FF MicroPlates in a closed<br />

container or in plastic bags on trays. Do not add<br />

additional moisture when incubating filamentous<br />

fungi.<br />
