3.2 <strong>Liquid</strong> medium culture<br />

<strong>Culture</strong> <strong>in</strong> LifeReactor © resulted <strong>in</strong> 0.35 new microshoots per explant,<br />

with an average length of 0.40 cm (Table 2). These small microshoots were<br />

<strong>in</strong>tensively hyperhydrized (macroscopically determ<strong>in</strong>ed) and small callus<br />

masses were present on the surface of the shoots and leaves (Figures 3b, 3c<br />

and 3d).<br />

<strong>Culture</strong>s on filter paper bridges <strong>for</strong>med 0.43 new microshoots per explant<br />

with an average shoot length of 0.32 cm (Table 2). Extensive callus<br />

<strong>for</strong>mation was noticed at the base of the explants (Figure 4d).<br />

<strong>Liquid</strong> culture of olive explants <strong>in</strong> Erlenmeyer flasks under agitation<br />

resulted <strong>in</strong> the highest number of new microshoots per explant (1.93) with an<br />

average shoot length of 0.40 cm (Table 2, Figure 4b). These microshoots<br />

suffered from the most serious hyperhydricity compared to the other systems<br />

tested.<br />

The novel TIS proved to be quite effective as far as the development of<br />

new microshoots <strong>in</strong> liquid media is concerned. The number of new<br />

microshoots per explant was 1.93 and shoot length was 0.95 cm <strong>in</strong> the case
