5 Using <strong>the</strong> <strong>Agilent</strong> <strong>7100</strong> <strong>Capillary</strong> <strong>Electrophoresis</strong> <strong>System</strong><br />

<strong>Capillary</strong> <strong>Electrophoresis</strong>, CE<br />

1 Make sure <strong>the</strong> buffer is filtered; use a 0.25 µm filter. For buffers with a high<br />

viscosity, a larger pore size, for example, 0.45 µm is also suitable.<br />

2 Fill <strong>the</strong> electrolyte bottle with buffer.<br />

For 500 mL bottles (part number 9300-1748), use a minimum of 100 mL <strong>to</strong><br />

make sure <strong>the</strong> frit is submerged in buffer. Use a maximum of 400 mL. For<br />

100 mL bottles (part number 5042-6478), use a minimum of 40 mL <strong>to</strong> make<br />

sure <strong>the</strong> frit is submerged in buffer. Use a maximum of 80 mL.<br />

3 Tighten <strong>the</strong> cap.<br />

Make sure <strong>the</strong> cap is tightened firmly, as pressure will be applied <strong>to</strong> <strong>the</strong><br />

bottle.<br />

4 Choose OK in <strong>the</strong> Change Bottles dialog box.<br />

Wait until <strong>the</strong> pressure has built up again, that is <strong>the</strong> instrument reaches<br />

<strong>the</strong> ready state.<br />

The right-hand bottle is <strong>the</strong> waste bottle. It is used <strong>to</strong> s<strong>to</strong>re buffer emptied<br />

from vials in <strong>the</strong> tray. Empty <strong>the</strong> bottle whenever you refill <strong>the</strong> electrolyte<br />

bottle, following <strong>the</strong> same procedure as described above. Empty <strong>the</strong> bottle<br />

frequently. The error message "Waste Bottle Full" appears if <strong>the</strong> waste bottle is<br />

full.<br />
