Setting Up a Test Sample Run<br />

Setting Up a Test Sample Run<br />

This instrument check demonstrates that <strong>the</strong> instrument is correctly installed<br />

and connected. Please use a fused silica capillary with 40 cm effective length<br />

and 50 µm inner diameter from <strong>the</strong> HPCE start-up capillary kit (part number<br />

G1600-68700) with <strong>the</strong> green alignment interface (part number G<strong>7100</strong>-60210).<br />

The instrument should be in CE mode.<br />

Load chemicals from <strong>the</strong> Installation Qualification Chemical kit (part number<br />

5063-6514) according <strong>to</strong> <strong>the</strong> table below. The Installation Qualification<br />

Chemical kit contains:<br />

• Buffer (20 mM borate, pH 9.3, 100 mL)<br />

• Test sample (4-hydroxyace<strong>to</strong>phenone 1 mM, 3 mL)<br />

• Conditioning solution (1.0 N sodium hydroxide, 100 mL)<br />

Choose <strong>the</strong> menu item Method > New method <strong>to</strong> prepare a method with <strong>the</strong><br />

parameters given below. Save <strong>the</strong> edited method under a new name. Choose<br />

<strong>the</strong> menu item RunControl > Sample Info… and enter <strong>the</strong> opera<strong>to</strong>r name,<br />

direc<strong>to</strong>ry and file name. Enter Vial location 7, enter sample name<br />

“1 mM pHAP” and click Run method <strong>to</strong> start <strong>the</strong> analysis.<br />

A typical electropherogram will show a significant peak at a migration time of<br />

around 3 minutes.<br />
