Vitis amurensis Rupr., distributed in the Northeast of China, is the most cold hardy grape<br />
species. It can tolerate temperatures as low as -40-50 C when fully cold acclimated. Our study<br />
showed that as the temperature drops, the cold hardy“Zuoshan-1”grapevine accumulates ABA<br />
quicker <strong>and</strong> in higher concentration than less cold hardy Vitis species. To better underst<strong>and</strong> the<br />
molecular mechanisms of ABA <strong>and</strong> its role in the grape cold hardiness, we constructed a<br />
suppression subtractive hybridization (SSH) cDNA library to isolate ABA inducible genes of<br />
“Zuoshan-1”. In a total of 214 r<strong>and</strong>omly picked clones, 167 were successfully sequenced from<br />
the ABA-induced library, from which 31 contigs were assembled. Blasting analysis revealed<br />
that14 contigs were homologous to Vitis or plant species while the other 17 contigs showed high<br />
homology to Escherichia coli, which were apparently resulted from E.coli contamination.<br />
Functional analysis indicated that the putative functions of these contigs were associated <strong>with</strong><br />
photosynthesis, cell defense or detoxification, <strong>and</strong> signal transduction.<br />
