A genetic analysis of berry quality traits in Spanish red wine grape<br />

The quality of wine is mainly dependent on the quality of the grape berry. Yield, berry ripening,<br />

berry size, acidity <strong>and</strong> sugar content, <strong>and</strong> phenolic maturity; expressed as total polyphenol index<br />

(TPI), color intensity (CI), tannin content, anthocyanin content (both extractable <strong>and</strong> total); are<br />

major traits associated <strong>with</strong> berry quality <strong>and</strong> aging of wine, respectively. These traits result from<br />

complex developmental processes influenced by genetic <strong>and</strong> environmental factors.A<br />

segregating population was constructed <strong>with</strong> 151 genotypes derived from a cross between two<br />

representative Spanish red wine varieties: Tempranillo <strong>and</strong> Graciano. Tempranillo is the most<br />

widely planted indigenous variety, present in 28 Spanish wine regions <strong>and</strong> is associated <strong>with</strong><br />

good quality wine. Graciano is a red Rioja variety used in blends <strong>with</strong> Tempranillo; Graciano has<br />

a later sprouting <strong>and</strong> maturation date, lower productivity, higher acidity, smaller berry size, <strong>and</strong><br />

higher phenolic content compared to Tempranillo. We have evaluated the population for three<br />

consecutive years. Phenotypic correlations among traits <strong>with</strong>in years; <strong>and</strong> between years <strong>with</strong>in<br />

traits were determined. Significant correlations were found between yield, CI, skin anthocyanins,<br />

total <strong>and</strong> extractable anthocyanins content, TPI, tannin <strong>and</strong> sugar content, <strong>and</strong> seed traits <strong>with</strong>in<br />

years. Simultaneously, parental genotypes <strong>and</strong> progeny were screened for 62 SSRs loci <strong>and</strong> 6<br />

AFLP primer combinations in a LI-COR IRD 4200 Sequencer. A genetic linkage map is being<br />

constructed for each parent using Joinmap 4.0. This will allow the identification of QTL<br />

responsible for traits of oenological interest in these representative Spanish red wine varieties.<br />
