Polyploid induction of mutation by using colchicines on tube seedlings of Victoria grape<br />

The unigemmius stem-segment of the tube seedlings of Victoria grape were soaked 24 hours <strong>and</strong><br />

48 hours by using colchicines aqueous solution of 0.05% <strong>and</strong> 0.1% concentration. The results<br />

showed that the best way for reduplicative efficiency was the third <strong>and</strong> fourth buds treated for 48<br />

hours in 0.05% colchicine <strong>and</strong> 24 hours in 0.1% colchicine, the doubling rates of primary<br />

generation cells were 33% <strong>and</strong> 31% respectively; The polyploid cell rate could increase about<br />

10% by subcultured the chimeras a generation. The polyploid cell rate could reach 75.8%-96.6 %<br />

by subcultured the chimeras four to six generations. Through root tip cell chromosome<br />

observation that it was a little effect to the division cycle of root-tip cells in terms of the tube<br />

seedlings <strong>with</strong> or <strong>with</strong>out illumination in short term., <strong>and</strong> the optimal sampling time of<br />

chromosome for observation was at 9:30 am.<br />
