Proteomic characterization of downy mildew-infected grapevine plants<br />

We have carried out a proteomic 2D analysis of Vitis vinifera leaves cv. Pinot Noir artificially<br />

infected <strong>with</strong> Plasmopara viticola (30000 sporangia per ml), <strong>and</strong> collected at 24, 48 <strong>and</strong> 96<br />

hours post-inoculation. Leaf tissues from 3 independent biological replicates were pooled.<br />

After testing different protocols for protein extraction, good quality 2D maps were obtained<br />

by performing protein extraction in a solubilizing/lysing solution containing 7M urea, 2M<br />

thiourea, 80mM citric acid, 1% C7Bz0 <strong>and</strong> 1x protease inhibitor cocktail. Gels (five per each<br />

sample) were scanned using a Bio-Rad VersaDoc 1000 imaging system <strong>and</strong> analyzed by<br />

PDQuest software (Bio-Rad) for spot detection, background subtraction <strong>and</strong> spot OD intensity<br />

quantification. After normalization <strong>and</strong> statistical analysis, about 100 spots differentially<br />

expressed were identified (fold-change ≥ 2, <strong>with</strong> p≤0.05). Identification by RP-HPLC-ESI-<br />

MS/MS analysis, revealed 72 unique proteins, 56 up-regulated <strong>and</strong> 16 down-regulated.<br />

Functional categories prevalently affected by infection were related to Calvin cycle <strong>and</strong><br />

glycolysis (28%), oxidative stress response (19%), defence response (12%), photosynthesis<br />

(9%) <strong>and</strong> protein folding (6%). A peculiar pattern in the number of modulated proteins over<br />

time was observed: 53 <strong>and</strong> 47 differentially expressed proteins were detected at 24 hpi <strong>and</strong> 96<br />

hpi, respectively, <strong>and</strong> only 14 at 48 hpi, none of which included in the “defence response”<br />

category, suggesting a temporary breakdown of the response, that will be the object of further<br />

investigations.<br />
