Chapter 7Maintaining the InstrumentDecontaminating the Sample BlockWhen to PerformMaterialsRequiredThe following procedure describes how to decontaminate the wells of a sample blockmodule. The procedure eliminates residual <strong>PCR</strong>-related products, includingfluorescent labeled TaqMan ® probes. Clean the sample block as often as needed.IMPORTANT! If you plan to use a cleaning or decontamination method other thanthe one in this manual, check with <strong>Applied</strong> <strong>Biosystems</strong> first to ensure that themethod will not damage the sample block module or the <strong>7900HT</strong> instrument.• 10% Sodium hypochlorite (bleach) solution• Isopropanol, 100 percent pure• 5/32 in hex key• Cotton swabsCHEMICAL HAZARD. Sodium hypochlorite (bleach) is aliquid disinfectant that can be corrosive to the skin <strong>and</strong> can cause skindepigmentation. Please read the M<strong>SDS</strong>, <strong>and</strong> follow the h<strong>and</strong>ling instructions. Wearappropriate protective eyewear, clothing, <strong>and</strong> gloves.CHEMICAL HAZARD. Isopropanol is a flammable liquid <strong>and</strong>vapor. It may cause eye, skin, <strong>and</strong> upper respiratory tract irritation. Prolonged orrepeated contact may dry skin <strong>and</strong> cause irritation. It may cause central nervoussystem effects such as drowsiness, dizziness, <strong>and</strong> headache, etc. Please read theM<strong>SDS</strong>, <strong>and</strong> follow the h<strong>and</strong>ling instructions. Wear appropriate protective eyewear,clothing, <strong>and</strong> gloves.Cleaning a LowDensity ArrayBlockThis procedure applies to the sample block on the <strong>7900HT</strong> <strong>System</strong> TaqMan LowDensity Array Upgrade.1. Wipe the surface of the sample block (grey aluminum) with 10% bleach.2. Wipe the sample block three times with distilled water.3. Wipe the sample block with isopropanol, then allow to air dry.WarningCircuitry <strong>and</strong> connections tothe instrument (Do Not Touch)Well A-1GR20284. Replace the block as explained in “Replacing the Sample Block” on page 7-10.5.
