A plate document is a virtual representation of a consumable (plate) usedto contain samples <strong>and</strong> reagents during a sequence detection run. During the run, thesoftware uses the plate document to record the operation of the instrument (thermalcycling <strong>and</strong> data collection parameters), organize <strong>and</strong> store the data gathered duringthe <strong>PCR</strong> process, store the of the run data, <strong>and</strong> record analysis <strong>and</strong> display options.Plate document templates have the same format as the <strong>SDS</strong> <strong>7900HT</strong> Documents(*.sds) but do not store raw data or bar codes.The objective of an absolute quantification experiment is to accurately determine theabsolute quantity of a single nucleic acid target sequence within an unknown sample.The results of an absolute quantification experiment are reported in the same units ofmeasure as the st<strong>and</strong>ard used to make them.One of the variant forms of a gene at a particular locus or location on a chromosomeThe process by which two variants of a single nucleic acid sequence present in apopulation of individuals' DNA samples is determined. Single nucleotidepolymorphisms (SNPs) comprise the most common variants that are assayed usingTaqMan reagents.A <strong>PCR</strong> amplified section of DNAThe Amplification plot displays data from real-time <strong>PCR</strong> for gene expression assaysafter signal normalization <strong>and</strong> multi-component analysis. The Amplification plotappears only in plate documents containing analyzed data from real-time runs. It isa 2D plot of normalized reporter fluorescence (for ∆R n ) values versus the cyclenumber. The ∆R n versus cycle Amplification plot is used for calculation thresholdcycle (C T ).A set of analysis results derived from a single raw data set associated with a singleplate document analyzed by the <strong>SDS</strong> software. The analysis session is persisted in thedatabase <strong>and</strong> can be retrieved by the <strong>SDS</strong> software. Multiple analysis sessions can becreated from the same plate document, however each analysis session must have aunique name. An analysis session does not have to be associated with a Study;however, if it has such a relationship, it can be associated with at most one Study atany given time.Binding a probe to the DNA or cDNA during the <strong>PCR</strong> processA set of routines, protocols, <strong>and</strong> tools for building software applications.
