5. Click on the row of the event of interest (or default to the first event), then click<br />

View to acquire the image of the highlighted event. <strong>iCys</strong> locates the image as<br />

follows:<br />

❍ If Field Image is selected:<br />

✧ The stage moves to the start of the scan field that contains the event of<br />

interest.<br />

✧ <strong>iCys</strong> moves the microscope objective to the focus position used during the<br />

original scan.<br />

✧ The scan field is rescanned using the setup from the original scan (number<br />

of lasers, PMTs, and so on).<br />

✧ The event is displayed and highlighted by a red box in the Field Image<br />

window.<br />

❍ If both Scan and Observation are selected:<br />

✧ After the appropriate scan field is rescanned, the stage moves so the event<br />

of interests is in the middle of the field of view through the eyepieces or<br />

color camera monitor.<br />

✧ <strong>iCys</strong> moves the microscope objective to the focus position used during the<br />

original scan.<br />

✧ To view the event through the eyepieces or camera monitor, turn the Light<br />

Path Selector Lever to the Observation position ( ).<br />

❍ If Observation is selected, the event is not rescanned.The following steps<br />

occur:<br />

✧ The stage moves so the event of interests is in the middle of the field of<br />

view through the eyepieces or color camera monitor.<br />

✧ <strong>iCys</strong> moves the microscope objective to the focus position used during the<br />

original scan.<br />

Note: When Observation is selected, <strong>iCys</strong> will turn the Brightfield on for<br />

viewing. Brightfield will be turned off when viewing an event with<br />

observation unchecked, or when closing the View Events window.<br />

In Observation-only mode, the event is not displayed in <strong>iCys</strong>.<br />

If Observation-only is selected, make sure the Light Path Selector lever is<br />

also in the Observation ( ) position.<br />

Note: To locate an event for which the Time value (in the View Events window)<br />

is different from the previous event’s value, and Scan is selected (with or<br />

without Observation), make sure the Light Path Selector lever is set to the<br />

Side Port position before selecting Next or View. This is required because<br />

the newly selected event is in a different scan area and will be rescanned.<br />
