6. For viewing, if the camera is on (as described in “Observing Samples With the<br />

Video Camera” on page 2-16) you can select the viewing mode by sliding the<br />

Light Path Selector knob as shown in Figure 2-10, “Light Path Selector Knob,” on<br />

page 2-15.<br />

7. Focus the sample using the microscope focus knob or, if available, the rocker<br />

switch and the fine focus knob on the Remote Control unit.<br />

8. After the hardware setup is complete, you can use the <strong>iCys</strong> application to view the<br />

samples on the microscope stage.<br />

Note: When using EpiFluorescence, you may cause photobleaching in the<br />

exposed samples. Photobleaching is caused by exposing fluorophores to<br />

light which, over time, destroys the fluorophores irreversibly and results<br />

in loss of fluorescence. The degree of photobleaching increases with the<br />

time and intensity of light exposure.<br />

When using the EpiFluoresence feature prior to scanning, minimize<br />

sample exposure and when possible scan in areas that have not been<br />

exposed to EpiFluorescence illumination.<br />

Observing Samples With the Video Camera<br />

<strong>iCys</strong> comes with a 3CCD camera module, EPIX PIXCI ® Imaging Board, and XCAP-<br />

Lite image viewing software. You can use the camera while observing to magnify and<br />

save images from the computer monitor.<br />

➣ To use the video camera:<br />

1. Turn on the color camera monitor.<br />

2. From your PC, click on XCAP for Windows to open the EPIX XCAP-Lite<br />

application.<br />

3. When <strong>iCys</strong> scans a sample or is moved from one event to another, the visual<br />

output is displayed on the camera monitor, if on, and on your PC, if XCAP-Lite is<br />

open.<br />

From XCAP-Lite, you can capture and save screenshots of desired images. See the<br />

online help for XCAP-Lite for more information.<br />
