Scanning Samples with <strong>iCys</strong><br />

➣ To scan samples with <strong>iCys</strong>, you need to make sure that the microscope is set as follows:<br />

1. Set the light path selector lever to side port ( ).<br />

Note: When set to observation ( ), scanning will not take place.<br />

2. Set the magnification selector knob to 1X magnification (the pushed in position).<br />

3. If your system has the optional EpiFluorescence feature, set the EpiFluorescence<br />

cassette to position 1.<br />

4. Set the condenser filter turret to position 5.<br />

Note: Position 5 of the condenser filter turret is used for brightfield and Forward<br />

Scatter and must be empty.<br />

5. Make sure the condenser is rotated in the down position (not rotated upwards).<br />

6. After the hardware setup is complete, you can use the <strong>iCys</strong> application to set up<br />

and scan as described in Chapter 3, “<strong>iCys</strong> Cytometric Analysis Software<br />

Application Overview.”<br />

Observing Samples with the Brightfield Source<br />

➣ To observe samples using the brightfield source:<br />

1. Set the light path selector lever to observation ( ).<br />

2. Turn on the following:<br />

❍ The TH4-100 Power Supply (shown in Figure 2-6, “Brightfield Illumination<br />

Assembly,” on page 2-10).<br />

❍ The brightfield ON/OFF switch (shown in Figure 2-9, “<strong>iCys</strong> Microscope<br />

Selected Components,” on page 2-13).<br />

You may want to set the Light Control Intensity knob on the TH4-100 to “Max”<br />

and use the Light Intensity Control knob on the main instrument to control the<br />

brightfield illumination.<br />

3. Select Brightfield from the <strong>iCys</strong> Instrument menu to open the brightfield shutter.<br />

For more information, see “Instrument Menu” on page 3-6.<br />

4. If your system has the optional EpiFluoresence feature, set the EpiFluorescence<br />

cassette to position 1.<br />

5. For viewing, if the camera is on (as described in “Observing Samples With the<br />

Video Camera” on page 2-16) you can select the viewing mode by sliding the<br />

Light Path Selector knob as shown in Figure 2-10:<br />
