Product Specific Information<br />
PowerVision+ Poly-AP IHC Detection systems are biotin-free, polymeric<br />
alkaline phosphatase (AP) conjugated systems, for the manual detection of<br />
tissue-bound mouse and rabbit antibodies.<br />
Components<br />
These detection systems all contain ready-to-use Post Primary Blocker and<br />
Poly AP Rabbit/Mouse.<br />
Antigen Retrieval Buffer<br />

Antigen Retrieval Buffers are intended for Heat Induced Epitope Retrieval<br />
(HIER) on formalin-fixed, paraffin-embedded tissue sections as part of an<br />
immunohistochemical procedure. HIER using an appropriate pH solution<br />
improves the staining of some antibodies by exposing epitopes within tissue<br />
that has been masked during fixation. A universal HIER technique suitable<br />
for all epitopes does not exist. A combination of different heating methods<br />
and epitope retrieval solutions may be used to optimize unmasking of<br />
antigens where this technique is recommended. HIER is not recommended<br />
for all antibodies. Optimum conditions for epitope retrieval should be<br />
validated by the user, as these are dependant upon tissue, fixation and/or<br />
primary antibody. The 10x solution is supplied in 1000 mL volume, sufficient<br />
to prepare 10 L of working solution.<br />
IHC/ISH Super Blocking<br />

IHC/ISH Super Blocking is specially formulated to stabilize antibodies and<br />
eliminate background encountered in immunohistochemistry (IHC) and in<br />
situ hybridization (ISH) procedures. The diluent contains complex protein<br />
and non-protein mixtures that eliminate background by blocking nonspecific<br />
protein-protein interactions. It is particularly recommended for<br />
staining of mouse tissues with mouse primary antibodies. It can also be used<br />
as a diluent for primary antibodies. If primary antibodies are diluted in this<br />
solution, a blocking step before primary antibodies can be omitted.<br />
Universal IHC Blocking/Diluent<br />

Research Use Only<br />
Universal IHC Blocking/Diluent can be used as a blocking reagent prior to<br />
antibody incubation, or as a diluent for antibodies. Pre-blocking should not<br />
be required if antibodies are diluted in this diluent.<br />
