B.6 MANNICH REACTION ON ISATIN<br />

The Mannich reaction consists in the condensation of ammonia or a<br />

primary or secondary amine, usually as the hydrochloride, with formaldehyde<br />

and a compound containing at least one hydrogen atom of pronounced<br />

reactivity. The essential feature of the reaction is the replacement of the active<br />

hydrogen atom by an aminomethyl or substituted aminomethyl group. The<br />

Mannich reaction is readily applied to isatins.<br />

Varma and Nobles 231 synthesized a series of Isatin-N-Mannich bases<br />

derived from isatin (Fig. B.1.6), 5-methyl and 5-bromoisatins as the active<br />

hydrogen component and dimethylamine, diethylamine, morpholine,<br />

piperidine, N-ethylcyclohexylamine, 3-azabicyclo [3.2.2] nonane and 3-azabicyclo<br />

[3.2.1] octane as the secondary amine moiety.<br />
