Chapter – 2 Microwave assisted simple and fast…..<br />

2.1 AIM OF CURRENT WORK<br />

Earlier it is already discussed that our group is involved in the synthesis<br />

of nitrogen containing heterocycles as they show good biological activity in<br />

different classes. Indole nucleus is known to possess different biological<br />

activities, thus it was decided to explore the chemistry involving isatin and 2methyl<br />

indoline moiety.<br />

Literature revealed that different types of N-alkylation reactions have<br />

been carried out on 2-methyl indoline and isatin, which include introduction of<br />

acetyl group, introduction of chloroacetyl group and further treatment with<br />

secondary amines, preparation of amide linkages and alkylation by means of<br />

one and two carbon chains. Looking to the reactivity of N-1 position in both<br />

the molecules, Mannich reaction had been carried out on 2-methyl indoline in<br />

chapter-1 and on isatin in chapter-3. Besides Mannich reaction, 2-methyl<br />

indoline and isatin can be suitable moieties for N-alkylation reactions.<br />

Literature survey revealed that very few N-Alkylation reactions have been<br />

carried out on 2-methyl indoline as well as on isatin moiety where two to three<br />

carbon atom chains have been introduced at N-1 position using basic<br />

conditions conventionally.<br />

There are many molecules available in the market existing as potent<br />

drug viz. Chlorpromazine, Fluphenazine, Prochlorperazine, Acetophenazine<br />

etc which are having excellent antipsychotic activity. These all molecules<br />

contain phenothiazine core moiety as well as long alkyl chain having two to<br />

three carbon atoms along with different substituents at the end of the alkyl<br />

chain on N atom. This inspired us to develop similar type of compounds using<br />

different alkyl chains as in above mentioned drugs but on indole nucleus.<br />

Thus we used 2-methyl indoline and isatin to develop new molecules. So this<br />

was one of the principal rational to carry out the work done in this chapter.<br />

Besides this, green chemistry and especially microwave assisted<br />

organic synthesis has drawn a remarkable attention of all chemistry<br />

researchers towards its advantages. There are many reactions reported<br />
