3.4.6 GENERAL PROCEDURE FOR THE PREPARATION OF DNJ-1301<br />

TO DNJ-1305<br />

To a stirred solution of 0.01 mole of 2-propylpentanohydrazide in 50 ml<br />

warm ethanol was added 0.01 mole of an appropriate N-Mannich base of<br />

isatin in 60 ml ethanol. Then 2–3 drops of glacial acetic acid were added and<br />

the reaction mixture was refluxed for 5 hours. The progress and the<br />

completion of the reaction were checked by silica gel-G F254 thin layer<br />

chromatography using toluene : ethyl acetate (7 : 3) as a mobile phase. After<br />

the reaction to be completed, the flask was cooled to afford a solid product.<br />

The solid residue was filtered, washed with cold ethanol and dried.<br />

Recrystallization was carried out from hot ethanol. (Physical data of the<br />

synthesized end products are summarized in the table 3.5.1)<br />

3.4.7 GENERAL PROCEDURE FOR THE PREPARATION OF DNJ-1401<br />

TO DNJ-1405<br />

It was again prepared by the method described by Chohan et. al. f To a<br />

stirred solution of 0.01 mole of 2-amino benzothiazole in 50 ml warm ethanol<br />

was added 0.01 mole an appropriate Mannich base of isatin in 60 ml ethanol.<br />

Then 2–3 drops of concentrated sulphuric acid were added and the reaction<br />

mixture was refluxed for 4 hours. The progress and the completion of the<br />

reaction were checked by silica gel-G F254 thin layer chromatography using<br />

hexane : ethyl acetate (6 : 4) as a mobile phase. After the reaction to be<br />

completed, the flask was cooled to afford a solid product. The solid residue<br />

was filtered, washed with cold ethanol, then with ether and dried.<br />

Recrystallization was carried out from hot ethanol. (Physical data of the<br />

synthesized end products are summarized in the table 3.5.2)<br />
