Content<br />

The practical work covers a range of laboratory exercises<br />

and common techniques used in biochemical and chemical<br />

laboratories. These techniques include estimation of<br />

disulphide and thiol groups in proteins, fluorescence<br />

spectroscopy, affinity chromatography, fractionation using<br />

ultracentrifugation, antibody labelling techniques, gel<br />

electrophoresis and enzyme kinetics.<br />

Preparation and characterisation of simple colloids.<br />

Potentiometric and conductimetric titrations of colloids to<br />

yield surface charge. Influence of electrolyte concentration<br />

and pH on surface charge. Micro-electrophoresis and the<br />

measurement of electrophoretic mobility (for colloids),<br />

which results in measurement of the zetapotential. The<br />

influence of zetapotential and electrolyte concentration (and<br />

type) on colloidal stability.<br />

Measurement of surface and interfacial tension.<br />

Determination of surface areas per molecule using Gibb's<br />

adsorption isotherm. Solution behaviour of surfactants -<br />

measurement of critical micelle concentration and the<br />

solubility (in water) of an oil-soluble molecule.<br />

Determination of surface area using adsorption from<br />

solution techniques. Influence of surFactant bn contact angle.<br />

Cleaning techniques and surface preparation. Flocculation<br />

rates. Emulsion polymerisation. Preparation of a paint.<br />

Polymer flocculation. Osmotic pressure. Dynamic light<br />

scattering. Porosimetry. Rheology and viscosity. The<br />

isoelectric point of a protein. Surface hydrophobicity.<br />

Preparation and characterisation of microemulsions.<br />

SC736 Research Skills, Part 1<br />

5 credit points 2 hours per week for 5 weeks Hawthorn<br />

This is a subject of the Graduate Diploma of Applied<br />

Science (Applied Colloid Science)<br />

Content<br />

Experimental design including the importance of the null<br />

result and 'blank' experiment, choice of expcrimental<br />

points, parameter variation and chernimetrics. The<br />

importance of prior preparation and choice of experimental<br />

variables. Literature searching skills and background theory.<br />

A major portion of this subject will be devoted to designing<br />

a project to be carried out either in SC538 or SC757.<br />

SC737 Research Skills, Part 2<br />

5 credit points 2 hours per week for 5 weeks Hawthorn<br />

This is a subject of the Graduate Diploma of Applied<br />

Science (Applied Colloid Science)<br />

Content<br />

Data manipulation and presentation, including statistics and<br />

methodology of data handling. Use of software packages in<br />

data manipulation. Presentation of data including an oral<br />

presentation involving work carried out either in SC538 or<br />

SC757.<br />

SC738 Minor Research Project<br />

7.5 credit points 4 hours per week for 5 weeks<br />

A minor research project carried out either at the student's<br />

place of work or at <strong>Swinburne</strong> and involving a closely<br />

Simple and complex rheology. Include complex rheology<br />
