Salinomycine was extracted fromformulations <strong>and</strong> feed by our original method – column chromatography on the column<strong>of</strong> silica gel G <strong>and</strong> elution with methanol. The content <strong>of</strong> salinomycin in eluates wasdetermined by spectrophotometric method at 520 nm with the vanilin reagens. Thedeveloped spectrophotometric method enables to determine the quantities <strong>of</strong> salinomycinin samples with detection limit <strong>of</strong> 0,5 mg/kg <strong>of</strong> feed. The recovery <strong>of</strong> spectrophotometricdetermination <strong>of</strong> salinomycin content in eluates was 83,8 <strong>and</strong> 103,5%, respectively. TheHPLC determination <strong>of</strong> salinomycine residues in prepared samples was accomplished bypost-column derivatisation with vanillin reagent <strong>and</strong> detection with VIS detector at 520nm. TSK ODS-120 T, 10µm (7,8 x 300mm) column <strong>and</strong> methanol-water-acetic acid(94:6:0.1, v/v/v) as mobile phase were used to determine SL. The recovery test wascarried out by adding st<strong>and</strong>ard solutions in concentrations from 10 to 60 ppm <strong>of</strong> feedsamples. The recovery rates for the HPLC method was over 96,3 <strong>and</strong> over 112,7%respectively. Detection limit <strong>of</strong> HPLC method was 15 µg/kg <strong>of</strong> feed.Key words: Salinomycine, spetrophotometry, liquid chromatography, poultry feedINTRODUCTIONCoccidiosis is an infective disease <strong>of</strong> the digestive tract which is most frequent withpoultry, causing a decrease in daily increment, prolonged fattening, poorer skinpigmentation, slower feed conversion <strong>and</strong> increased mortality. The disease is caused byProtozoas from the genera <strong>of</strong> Eimeria, Isospora <strong>and</strong> Cryptospora, <strong>and</strong> it is manifested bydamaging the intestine epithelial cells, less frequently the bile duct <strong>and</strong> renal tubuli [3,10].Worldwide intensive study <strong>of</strong> new products which would be added to poultry feed toprevented the coccidiosis, methods for their determination in premixes, finishedmixtures, as well as methods for the determination <strong>of</strong> their residues in tissues <strong>of</strong> treated276
