747 Effect of Dietary Supplementation of Tinospora Cordifolia on Metabolic Profile, Antioxidant Status and Immune Response in Lambs under Semi-Intensive System D. Nagalakshmi 1 , K. The reference lambs received the basal concentrate mixture (no herb) and the experimental lambs received 60, 90 and 120mg/kg BW of Tinospora cordifolia, respectively for a period of 90days. The blood haematological, biochemical and oxidative enzyme activity measured at 0, 45 and 90d indicated higher (P

Having generated all the data, genetic and environmental parameters, such as means and variances of genetic, underlying phenotypic and threshold phenotypic were calculated for each generation. The underlying phenotypic data were analysed by ASREML version 2.00a with animal model. The threshold phenotypic data were analysed by MATVEC using threshold model. Results: Having calculated genetic parameters for all generations, it was found that genetic and phenotypic means increased as well as threshold mean. The results showed that response to auto selection depended on dam distribution for the threshold phenotype. Furthermore, heritability remained constant across generations and there was no significant difference in compare with the base generation. Analysing threshold data resulted that corresponding heritabilities at theoretical 0.10, 0.25 and 0.50 for underlying phenotype in base population were estimated to be 0.0643, 0.1697 and 0.2807, respectively. Conclusions: Auto selection increases the genetic, phenotypic and threshold means, but had no effect on heritability. However, analysis of threshold data with threshold model resulted in under-estimate heritabilities compare with their corresponding underlying heritabilities. However, little is known about the use of cell count techniques and its usefulness in sheep, mainly in relation to Lacaune dairy sheep. The present study analyzed 200 samples of milk collected from the halves of 100 Lacaune sheep in different phases of lactation. The first aliquot was used in automatic cell counts by means flow citometry; the second aliquot was used in direct microscopic counts using methyl green-pyronin Y and Rosenfeld; the third aliquot was used in bacteriological examination. From the 200 samples analyzed, 14 were positive at the bacteriological examination, 2 samples were contaminated and 184 were negative. Bacteria isolated and identified were: Staphylococcus hyicus (n=1); Staphylococcus Kloosii (n=7); Staphylococcus haemolyticus (n=5); and Staphylococcus epidermidis (n=1). Results for automatic cell counts were 0.15 and 0.113x10 6 ; for microscopic cell counts using methyl greenpyronin were 0.126 and 0.320 x10 6 ; and microscopic counts using Rosenfeld were 0.127 and 0.851 x10 6 , for samples positive and negative in the bacteriological examination, respectively. Results for cell counts showed significant differences between the groups of positive and negative animals only when methyl green-pyronin Y and Rosenfeld were used, making it necessary to be very careful in the choice of the adequate technique for the detection of mammary infection. Fecal samples were collected weekly until lambs were 12 weeks old, a last sample was collected at the 16th week. Oocysts were quantified in each sample according to Gordon and Whitlock’s technique modified by Ueno and Gonçalves (1998). Eimeria species were identified according to morphological characteristics of oocysts and sporocysts (Levine, 1985; Reginsson and Richter, 1997). The oocysts excretion curves were similar for all treatments, but not the time in which each event happened. Control group lambs had a gradual rise in oocysts excretion after the 3 rd week of age. Two peeks occurred until the 12 th week of age, one at 7 th (125985 OOPG) and other at 10 th week (101657 OOPG). For the other groups this rise began between the 3 rd and 6 th weeks. Groups 2 and 4 also had 2 peaks, respectively, at the 8 th (49708 OOPG) and 12 th (47933 OOPG), and at 9 th (220521 OOPG) and 12 th (108416 OOPG) weeks of age. Group 1 had only 1 peak at the 10 th week (119654 OOPG). After the 10 th week of age oocysts output of all groups gradual and continuously reduced, maintaining low levels until the 16 th week of age.
