INTERNATIONAL SOCIETY FOR THE STUDY OF HARMFUL ALGAE12 th <str<strong>on</strong>g>Internati<strong>on</strong>al</str<strong>on</strong>g> <str<strong>on</strong>g>C<strong>on</strong>ference</str<strong>on</strong>g> <strong>on</strong> <strong>Harmful</strong> <strong>Algae</strong>, Copenhagen, Denmark, 4-8 September 2006The growth capabilities of thetoxigenic diatom species Pseud<strong>on</strong>itzschiacuspidata, isolated fromthe Juan de Fuca Eddy regi<strong>on</strong>during Fall surveys of the ECOHAB-PNW project in 2004 and 2005,were examined in n<strong>on</strong>-axenic, semic<strong>on</strong>tinuous,batch cultures enrichedwith 40 µM nitrate, 40 µMamm<strong>on</strong>ium or 20 µM of urea as thesole nitrogen source. Experiments,c<strong>on</strong>ducted at both high (120µmol·m -2·s -1 ) and low (40 µmol· m -2·s -1 ) photosynthetic phot<strong>on</strong> fluxdensities (PPFDs), dem<strong>on</strong>stratethat P. cuspidata grew significantlyfaster at the higher PPFD, butshows no preference for <strong>on</strong>enitrogen source over the other ateither PPFDs. Exp<strong>on</strong>ential growthrates (determined using cellabundance over time) andparticulate domoic acid c<strong>on</strong>tent(using ELISA) did not significantlydiffer as a functi<strong>on</strong> of the nitrogengrowth substrate. In c<strong>on</strong>trast tomost previous studies using othercultured Pseudo-nitzschia species,the particulate DA per cell averaged60% greater during exp<strong>on</strong>entialgrowth compared to stati<strong>on</strong>arygrowth of P. cuspidata, regardlessof nitrogen source or PPFD. Theseresults dem<strong>on</strong>strate the capability ofthis diatom to grow equally well <strong>on</strong>both oxidized and reduced forms ofnitrogen, although our fieldobservati<strong>on</strong>s have generally foundgreatest cell abundances in thenitrate-rich upwelled waters of theJuan de Fuca Eddy.PO.13-65Cyanobacterial toxins in thelakes located in the Riga City andits surroundingsSessi<strong>on</strong>: PO.13 - Regi<strong>on</strong>al eventsMaija Balode, Ingrida Purina, IevaBarda, Solvita StrakeLatvian Institute of Aquatic Ecology, RIGA,LatviaThe coastal z<strong>on</strong>e of the lakeslocated in the Riga City and itssurroundings are densely inhabitedand widely used for recreati<strong>on</strong>,fishing activities and as drinkingwater source for Riga City. Due tostr<strong>on</strong>g anthropogenic impact,intensive blooms of potentially toxicalgae in the lakes are observedevery summer, however no regularanalysis of algal toxins has beencarried out. In this study weattempted to quantify the toxinc<strong>on</strong>centrati<strong>on</strong>s in different stages ofcyanobacterial blooms in 2002-2005. Samples for cyanobacterialtoxin analysis were collected fromalgae surface accumulati<strong>on</strong>s.Presence of toxins was detected byprotein phosphatase inhibiti<strong>on</strong>assay (PPIA), enzyme linkedimmunosorbent assay (ELISA) andHPLC. The most frequentlyoccurring species in all sampleswere Microcystis spp., Anabaenaspp. and Aphanizomen<strong>on</strong> flosaquae,composing 50 -95% of thetotal phytoplankt<strong>on</strong> biomass. Thehighest c<strong>on</strong>centrati<strong>on</strong>s ofmicrocystins were detected in thelakes Mazais Baltezers and LielaisBaltezers, ranging from 300- 1600ng/mg dw during the maximum ofthe cyanobacterial bloom.
